Data on the aquatic toxicity of Glycerol monomyristate (CAS 27214-38-6) are available for aquatic invertebrates and algae. In order to assess the acute toxicity to fish and the toxicity to microorganisms read-across to the structurally related substances Glycerides, C12-18 mono- and di- ( (CAS 91052-49-2), Glycerides, C12-18 di- and tri- (CAS 91744-28-4) and Docosanoic acid, ester with 1,2,3-propanetriol (CAS 77538-19-3) was conducted to fulfil the standard information requirements. In accordance with Article 13 (1) of Regulation (EC) No 1907/2006, "information on intrinsic properties of substances may be generated by means other than tests, provided that the conditions set out in Annex XI are met.” In particular for aquatic toxicity, information shall be generated whenever possible by means other than vertebrate animal tests, which includes the use of information from structurally related substances (grouping or read-across). According to Annex XI, Item 1.5, of Regulation (EC) No 1907/2006 substances can be considered to be similar if their physicochemical, toxicological and ecotoxicological properties are likely to be similar or follow a regular pattern as a result of structural similarity.

The target and source substances are esters of a tri-functional alcohol (glycerol) with one or more carboxylic (fatty) acid(s) chain(s). The alcohol moiety (glycerol) is common to allanalogue substances.The fatty acid moiety comprises linear saturated carbon chain lengths from C12-C18 (even-numbered) and C22 bound to the alcohol, resulting in mono-, di-, and triesters.The target and the source substances are characterised by comparable physico-chemical properties and an analogue environmental fate. Please refer to the analogue justification for more details on the read-across approach.

 

The available studies on the acute toxicity of the source substance to fish determined no detrimental effects on fish. The determined LL50 (96 h) forGlycerides, C12-18 mono- and di- (CAS 91052-49-2) was > 2.7 mg/L (measured). For the source substance of Docosanoic acid, ester with 1,2,3-propanetriol (CAS 77538-19-3) a LC50 (96 h) > 10000 mg/L (nominal concentration) was reported.

 

The available study on the toxicity of the target substanceGlycerol monomyristate to aquatic invertebrates determined noacute toxicity to Daphnia magna (EC50 (48 h) > 0.69 mg/L; measured, arithm. mean). In the available study on the toxicity ofGlycerol monomyristate to aquatic algae anErC50 (72 h) >0.28 mg/L (meas. arith. mean) was determined (ErC10 (72 h) >0.28 mg/L (meas. arith. mean)).

Data on the toxicity to microorganisms are available for the source substance Glycerides, C12-18 di- and tri- (CAS 91744-28-4). Adverse effects of the substances to microorganisms were not observed.

The available acute toxicity data on the target and the source substances demonstrate that acute toxic effects of the target substance Glycerol monomyristate (CAS 27214-38-6) are not likely.
