Test temperature:
The column temperature during the study was monitored every 2 minutes by a data logger. A mean value of 21.1 °C at pH 5.5 and 20.8 at pH 7.5 was obtained for the temperature during the course of the study.

Details on study design: HPLC method:
TEST SYSTEM Analytical HPLC-column (based on silica matrix), coupled to DAD, packed with a cyanopropyl solid phase containing non-polar and polar moieties. Methanol : KH2PO4 buffer (pH 7.5) (70 :30 v/v) and Methanol : KH2PO4 buffer (pH 5.5) (70 :30 v/v) were used as mobile phase.

Reasons for the selection The selection of the test system is based on the guidelines. HPLC coupled to DAD is a useful method for estimation of the adsorption coefficient KOC. The KOC indicates the binding capacity of the test item on soil or sewage sludge and allows comparing the adsorption behaviour of different chemicals. The test item and reference items will be detected by DAD.

Results of Test Item Analysis at pH 5.5

The table below contains the retention time data and calculated log k’ values for Disperse Orange 29 at pH 5.5. Two individual log KOC values for the test item were derived under consideration of the first and second calibration. Finally, a mean value was calculated from these individual log KOC value for soil and for sewage sludge.
Individual log KOC values of 5.62 ± 0.37 (95 % CI 4.71 to 6.54) and 5.68 ± 0.38 (95 % CI 4.76 to 6.60) were obtained with both calibrations for the soil and log KOC values of 5.89 ± 0.55 (95 % CI 4.48 to 7.30) and 5.97 ± 0.57 (95 % CI 4.51 to 7.42) for the sewage sludge. The individual values did not differ more than ± 0.25 log KOC values. Based on these data a mean value of 5.65 was derived for soil and 5.93 for sewage sludge. Since the mean retention time for the test item was 9.73 min at pH 5.5, these values are extrapolated because the retention time of the test item was higher than that of the reference item with the highest log KOC.

Results of Test Item Analysis at pH 7.5

The table below contains the retention time data and calculated log k’ values for Disperse Orange 29 at pH 7.5. Two individual log KOC values for the test item were derived under consideration of the first and second calibration. Finally, a mean value was calculated from these individual log KOC value for soil and for sewage sludge.
Individual log KOC values of 3.52 ± 0.16 (95 % CI 3.14 to 3.90) and 3.52 ± 0.15 (95 % CI 3.14 to 3.89) were obtained with both calibrations for the soil and log KOC values of 3.51 ± 0.21 (95 % CI 2.98 to 4.04) for the sewage sludge. The individual values did not differ more than ± 0.25 log KOC values. Based on these data a mean value of 3.52 was derived for soil and 3.51 for sewage sludge.

Remarks:
Calculated values for log KOC of the test item did not differ by more than 0.25 between injections. The variation in temperature was between ± 1 °C during the course of measurements. A signal-to-noise ratio > 3 was provided for all substances.
Conclusions:
The adsorption coefficient log Koc of the test substance was determined to be 5.65 for soil and 5.93 for sewage sludge at pH 5.5 (21.1 °C) and 3.52 for soil and 3.51 for sewage sludge at pH 7.5 (20.8 °C).
Executive summary:

A study was conducted to determine the adsorption coefficient (Koc) of the test substance (94.8% purity) on soil and on sewage sludge using high performance liquid chromatography (HPLC) according to OECD Guideline 121 and EU Method C.19. Two pH values were used since the test substance is ionisable in the relevant pH range of 5.5 to 7.5. The temperature was controlled (21.1 °C for the buffer 5.5 and 20.8 °C for the buffer 7.5, mean value). A methanol : KH2PO4(pH 5.5) (70 : 30 v/v) and a methanol : KH2PO4 buffer (pH 7.5) (70 : 30 v/v) were used as mobile phases. A 10mg/L sample of the test substance was analysed threefold via HPLC on a cyanopropyl (CN-) column with DAD detection. Calibrations were performed using eight reference items for soil and seven reference items for sewage sludge with known log Koc values (given by the guidelines). The reference items were measured twice, once before and once after the measurement (double injection of one replicate) of the test item. Dead time of the HPLC-system was determined with sodium nitrate. Two individual log Koc values for soil and for sewage sludge were derived under consideration of the first and second calibration at each pH value. Finally, a mean value for soil and for sewage sludge was calculated from these individual log Koc values. The log Koc values were determined to be 5.65 for soil and 5.93 for sewage sludge at pH 5.5 (21.1 °C). Values at pH 7.5, extrapolated because the retention time of the test item was higher than that of the reference item with the highest log Koc, were equivalent to 3.52 for soil and 3.51 for sewage sludge (20.8 °C) (Landsberg, 2016).
