Description of key information

Reliable skin and eye irritation studies in rabbits were performed (similar to OECD 404 and 405 respectively, non-GLP). 
DL-Alpha-Tocopheryl acetate is not irritating to the skin and eyes.

Additional information

In a skin irritation study, performed according to OECD guideline 404, 3 rabbits (2 males, 1 female) were exposed to DL-Alpha-Tocopheryl acetate. Erythema and edema were scored at 24, 48, and 72 hours and the mean score was 0.0 at all time points. The test substance was not a skin irritant under the conditions of this study (BASF AG, 1989). This finding was supported in another study in which the test substance was applied under occlusive conditions (DSM, 1981).

In an eye irritation study, performed according to OECD guideline 405, 3 rabbits (1 male, 2 females) were exposed to DL-Alpha-Tocopheryl acetate.

Opacity, iris, redness and swelling were scored at 24, 48, and 72 hours and the mean scores were 0.0, 0.0, 0.2, and 0.0 respectively.

DL-Alpha-Tocopheryl acetate

was not an eye irritant under the conditions of this study (BASF AG, 1989). This finding was also noted in another supporting study (DSM, 1981).

Justification for classification or non-classification

Findings do not meet the criteria for classification as skin or eye irritant according to EU Directive 67/548/EEC and EU classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008.
