Details on test material:
Name of the test substance used in the study report: Nitrobiphenyl (Reg. Nr. 363 986)
Degree of purity: 99.5%

Method

Target gene:
his, trp
Species / strain
Species / strain / cell type:
S. typhimurium TA 1535, TA 1537, TA 98, TA 100 and E. coli WP2
Metabolic activation:
with and without
Metabolic activation system:
Aroclor-induced rat liver S-9 mix
Test concentrations with justification for top dose:
20 µg - 5000 µg/plate (Standard Plate Test; TA 1535, TA 98, E. coli)
15.625 µg - 5000 µg/plate (Standard Plate Test; TA 100, TA 1537)
15 µg - 250 µg/plate (Preincubation Test; Salmonella strains)
4 µg - 2500 µg/plate (Preincubation Test E. coli)
Vehicle / solvent:
Due to the insolubility of the test substance in water, DMSO was selected as the vehicle, which had been demonstrated to be suitable in bacterial reverse mutation tests and far which historical control data are available.

Additional information on results:
An increase in the number of his+ or trp+ revertants was not observed in the standard plate test or in the preincubation test either without S-9 mix or after the addition of a metabolizing system.
Remarks on result:
other: all strains/cell types tested
Remarks:

Any other information on results incl. tables

Test substance precipitation was found from about 250 µg - 500 µg/plate onward.

A bacteriotoxic effect was observed in the standard plate test with Salmonella strains depending on the strain and the test conditions from about 100 µg - 500 µg/plate onward.

Applicant's summary and conclusion

Conclusions:
Interpretation of results (migrated information):
negative

According to the results of the present study, the test substance is not mutagenic in the Salmonella typhimurium/Escherichia coli reverse mutation assay under the experimental conditions chosen here.
