Description of key information

The 48 hr EC50 to aquatic invertebrates was calculated to be 0.92 mg/L for a representative C8 structure. For C12 and C16 structures no effects were predicted at saturation – which supports the experimental data for the C12 stream.

Additional information

Two studies were available for the stream 97280-83-6 dodecene, branched (predominant carbon number of C12). 

Huntingdon Life Sciences (2009a) report that due to the low solubility of the test substance a WAF method was used to prepare the stock solution. The WAF was prepared by adding 100 mg of the test item to the test media and stirring overnight. This solution was used as the single exposure concentration in a 48 hour static test. Results are reported based on the nominal loading rate. No effects were observed at this concentration. The 48 hour EC50 was therefore determined to be > 100 mg/L WAF. This value have been used in the PBT assessment of this category.

Infracor (2000) provide no detail on how the stock solution was prepared, however the exposure concentrations were confirmed by analysis at the start and end of the test. The highest exposure concentration which could be produced was 3.1 mg/L (geometric mean). No effects were observed at any test concentrations, so the 48 hour EC50 was therefore determined to be > 3.1 mg/L. 

In the absence of measured data for the other category members, QSAR estimates (ECOSAR, part of EPISuite V4.0) were used to predict toxicity based on representative members of the category. The 48 hr EC50 to aquatic invertebrates was calculated to be 0.92 mg/L for a representative C8 structure. Classification would therefore be based on this calculated data and this would result in a classification of R50/53 (DSD) and Acute and Chronic Category 1 (CLP) as the lowest predicted EC50 is 0.92 mg/L for Daphnia and the predicted log Kow for this stream is >4.

For C12 and C16 structures no effects were predicted at saturation – which supports the experimental data for the C12 stream.
