Activated sludge was obtained as the test inoculum from a sewage plant treating predominantly domestic sewage (Nanjing Tiebei Sewage Treatment Plant). On return to the laboratory, coarse particles were removed by filtration through a fine sieve and the sludge aerobic will be kept thereafter for 5-7 days at the test temperature.

The vessels will be allowed to reach the desired temperature and appropriate vessels will be inoculated with prepared activated sludge to give a concentration of suspended solids at 30 mg/L.
Addition of Test Substance – appropriate amount of test substance will be added into test bottles of “test” (bottle 1&2) and “Inhibition control” (bottle 6).
Addition of Reference Substance – 10 mL of the reference control stock solution of Sodium benzoate (1000 mg/L) will be added into test bottles of “procedure control” (bottle 5) and “Inhibition control” (bottle 6) to give a final concentration of 40 mg/L, respectively.
Addition of Inoculums – 1.875 mL of Inoculums suspended solids will be added into “test” (bottle 1&2), “inoculum control” (bottle 3&4), “procedure control” (bottle 5) and “Inhibition control” (bottle 6) to give a concentration of 30 mg/L.
Basal Salts Medium (BSM) will be added to every test bottle to get a final volume of 250 mL.
The equipment will be assembled, the stirrer started, the equipment checked that it is air-tight, and the measurement of oxygen uptake will be started (Time zero). Carbon dioxide produced metabolically by the bacteria is chemically bound by the sodium hydroxide solution contained in the seal cup in the bottle. Usually no further attention is required other than taking the necessary readings and making daily checks to see that the correct temperature and adequate stirring are maintained. According to the readings of automatic respirometer, a continuous record of oxygen uptake is obtained so that the 10-d window is easily recognized.
The oxygen uptake will be calculated from the readings taken at least daily, using the methods given by the manufacturer of the equipment.
At the beginning and the end of incubation, normally 28 days, the pH of the contents of the flasks will be measured
