PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: 0.0503 g test item was weighed and dissolved in 500 mL of test water by magnetic stirring for 24 hours. After magnetic stirring, undissolved test item was observed in the solution, the solution was filtered through a 0.45 μm filter membrane and the filtrated solution was used to dilute other test solutions. Pipette 25.0, 40.0, 67.5, 107.5 and 175.0 mL of the filtrated solution and make up to 250 mL with test water. The solutions were mixed thoroughly, resulting in test solutions at 10%, 16%, 27%, 43% and 70% of the saturated solution prepared at 100 mg/L.

During the 48-hour exposure period of the first definitive test, no immobilization and abnormal behavior or appearance was observed in the control group and all test groups.
During the 48-hour exposure period of the second definitive test, no immobilization and abnormal behavior or appearance was observed in the control group and the test groups of 20%, 30%, 45%, 67% of the saturated solution prepared at 100 mg/L, the daphnids exposed to the test group of 100% of the saturated solution prepared at 100 mg/L showed abnormal behavior such as hypoactivity and immobilization.
Results with reference substance (positive control):
EC50-24 h of potassium dichromate to Daphnia magna was 1.44 mg/L, and the 95% confidence limits was 1.23 ~ 1.69 mg/L. The results met the recommended criterion in OECD Guideline No. 202: EC50-24 h of potassium dichromate is within the range of 0.6 mg/L to 2.1 mg/L.
