Sotck solution of the test substance: A stock solution of 995 mg/L (based on the active component) is prepared by dissolving 565.3 mg of the test substance in 200 mL deionised water.

Test concentrations: the following test concentrations - made up in a geometric series at a concentration ratio of 2.0 - are prepared by appropriate dilution of the stock solution with algal medium: 0 (control) - 0.005 - 0.010 - 0.020 - 0.040 - 0.080 - 0.160 and 0.319 mg/L (nominal concentration based on the active component).

Volume of test solution: 100 mL of test solution in each erlenmeyer.

Light intensity: Continious uniform illumination is provided by Philips TL-33 lamps yielding 4200 Lux at the surface of the test solution as measured with a spherical collector.

Species: Selenastrum capricornutum

Method of cultivation: the culturing of Selenastrum capricornutum for use in the alga growth inhibition test is carried out according to the procedure outlined in the Annex of OECD Guideline 201. The pre-culture of Selenastrum capricornutum which is used for the inoculation of the test cultures is incubated under the conditions of the test. After an incubation period of 5 days, the cell density of the pre-culture is 2.0x10E6 cells/mL. Microscopic observation reveals that the pre-culture contains no deformed or abnormal algal cells.

Initial cell concentration: the initial cell concentration of Selenastrum caprucornutum in the test cultures is 1.0x10E4 cells/mL.

Shaking: In order to keep the algae in suspension and to facilitate transfer of CO2, the test erlenmeyer are shaken manually three times per day.
