PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Preliminary media trial results:
Visual observations made on the centrifuged test samples showed they were hazy homogenous dispersions, as such the use of centrifugation as a means to remove the undissolved test item present was considered inappropriate. Whilst the filtered samples yielded results of less than the LOQ, determined to be 0.029 mg/L, this does not infer that no test item was in solution, just that any which was present was so at a concentration of less than 0.029 mg/L.
- Method: Based on the preliminary trial the test item was prepared using a saturated solution method of preparation at an initial loading rate of 50 mg/L, stirred for a period of 24 hours prior to the removal of any undissolved test item by filtration through a 0.2 μm Sartorius Sartopore filter (first approximate 1 liter discarded) to give a 100% v/v saturated solution of the test item. The test water used for both the range-finding and definitive tests was the same as that used to maintain the stock fish.

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water:
The test water used for both the range-finding and definitive tests was the same as that used to maintain the stock fish.
Laboratory tap water was dechlorinated by passage through an activated carbon filter (Purite Series 500) and partly softened (Elga Nimbus 1248D Duplex Water Softener) giving water with a total hardness of approximately 140 mg/L as CaCO3. After dechlorination and softening the water was passed through a series of computer controlled plate heat exchangers to achieve the required temperature.

Chemical analysis of the 100% v/v saturated solution fresh test solution at 0 hours and 24-Hour old test media showed measured test concentrations of less than the limit of quantification (LOQ) of the analytical method employed was obtained which was determined to be 0.0048 mg/L. Based on this information, a single test concentration of 100% v/v saturated solution was selected for the definitive test. The test was considered to be valid given that none of the control fish died or showed signs of stress during the test and that the oxygen concentration at the end of the test was ≥60% of ASV (6.1 mg O2/L) in the control and test vessels.

SUMMARY

1. Introduction

A study was performed to assess the acute toxicity of the test item to rainbow trout (Oncorhynchus mykiss). The method followed was designed to be compatible with the OECD Guidelines for Testing of Chemicals (1992) No 203, "Fish, Acute Toxicity Test" referenced as Method C.1 of Commission Regulation (EC) No. 440/2008.

2. Methods

Information provided by the Sponsor indicated the water solubility of the test item to be 5.8 x 10-5 mg/L.

Preliminary solubility work conducted indicated that it was not possible to obtain a testable solution of the test item using traditional methods of preparation e.g. ultrasonication and high shear mixing. A preliminary media preparation trial indicated that a saturated solution method of preparation was appropriate for this test item.

Seven fish were exposed to an aqueous solution of the test item, at a single concentration of 100% v/v saturated solution for a period of 96 hours at a temperature of approximately 14 ºC to 16 °C under static test conditions. The test item solution was prepared by stirring an excess (100 mg/L) of test item in test water using a propeller stirrer at approximately 1500 rpm for 48 hours. After the stirring period any undissolved test item was removed by filtration (0.2 μm Sartorius Sartopore filter, first approximate 1 litre discarded in order to pre-condition the filter) to produce a saturated solution of the test item with a nominal concentration of 100% v/v saturated solution. The number of mortalities and any sub-lethal effects of exposure in each test and control vessel were determined 1, 3 and 6 hours after the start of exposure and then daily throughout the test until termination after 96 hours.

3. Results
