There is no study available investigating the short-term toxicity of Potassium (Z)-N-methyl-N-(1-oxo-9-octadecenyl) aminoacetate (CAS 76622-74-7) to fish. In accordance to Regulation (EC) No 1907/2006 Annex XI, 1.5 a read-across to the structurally related source substances Sodium N-methyl-N-(1-oxo-9-octadecenyl)aminoacetate (CAS 3624-77-9) with sodium hydroxide and (Z)-N-methyl-N-(1-oxo-9-octadecenyl)glycine (CAS 110-25-8) was conducted. Based on the high degree of similarity between the structural and physico-chemical properties of the target and source substances, the source substances are considered suitable representatives for the evaluation of the short-term toxicity of the target substance to fish. The read-across approach is justified in detail within the analogue justification in IUCLID section 13.

Both studies were conducted according to the OECD guideline 203 and GLP.

The first study investigated the short-term effects of the source substance Sodium N-methyl-N-(1-oxo-9-octadecenyl)aminoacetate (CAS 3624-77-9) to freshwater fish. D. rerio was exposed to the nominal test item concentrations of 0.1, 0.25, 0.5, 1.0, and 2.0 mg/L for 96 h. The test item concentrations were not analytically verified. The obtained LC50 (96 h) was 0.37 mg/L.

In the second study the acute toxicity of (Z)-N-methyl-N-(1-oxo-9-octadecenyl)glycine (CAS 110-25-8) to fish was investigated in a limit test using a flow-through test design. D. rerio was exposed to a nominal concentration of 0.43 mg/L test item for 96 h in flow-through conditions. The test item concentration was analytically verified at the start and end of the test as well as every 24 h. The analytical measurements show 91 – 107% of the nominal test concentrations at test start and every 24 h until test end. Therefore, the nominal concentration was used for the determination of the results. After 96 h no effects were observed and the obtained LC50 (96 h) was > 0.43 mg/L.

Based on the available results from the structurally related source substances (in accordance with Regulation (EC) No 1907/2006 Annex XI, 1.5), which are characterized by a similar ecotoxicological profile and comparable structure, an LC50 (96 h) of 0.37 mg/L for fish is retained as effect concentration for the toxicity of the target substance Potassium (Z)-N-methyl-N-(1-oxo-9-octadecenyl) aminoacetate (CAS 76622-74-7) to fish.
