Short-term toxicity to fish

Data for the target chemical substance 7-[[2-[(aminocarbonyl)amino]-4-[[4-chloro-6-[[4-[vinylsulphonyl]phenyl]amino]-1,3,5- triazin-2-yl] amino]phenyl]azo]naphthalene-1,3,6-trisulphonic acid, sodium salt (CAS No. 94158 -80 -2) and read across substance were reviewed to summarize the following information:

Based on the QSAR prediction done using the EPI Suite ECOSAR version 1.10, the 96 hours LC50 was estimated to be 15900.743 mg/L on fish for substance7-[[2-[(aminocarbonyl)amino]-4-[[4-chloro-6-[[4-[vinylsulphonyl]phenyl]amino]-1,3,5-triazin-2-yl]amino]phenyl]azo]naphthalene-1,3,6-trisulphonic acid, sodium salt with mortality effects.

Whereas read across data from experimental study indicate that the test dye Direct Yellow 50 was determine to access the toxic effects in aquatic environment.

During 96 hrs study the lethal concentration (LC50) for test substance was found to be 180 mg/l in freshwater and static condition for fish Pimephales promelas on the basis of mortality effects.

Thus based on the result it is concluded that the test substance is non toxic to fish. (Dyes and the Environment - Reports on Selected Dyes and Their Effects, American Dye Manufacturers Institute:123 p.; 1973)

Thus based on the above available information it is concluded that the test substance 7-[[2-[(aminocarbonyl)amino]-4-[[4-chloro-6-[[4-[vinylsulphonyl]phenyl]amino]-1,3,5- triazin-2-yl] amino]phenyl]azo]naphthalene-1,3,6-trisulphonic acid, sodium salt (CAS No. 94158 -80 -2) is non toxic to fish and thus not consider for the further classification as per the CLP regulation.

Toxicity to aquatic algae and cyanobacteria

Various predicted data for the target chemical substance7-[[2-[(aminocarbonyl)amino]-4-[[4-chloro-6-[[4-[vinylsulphonyl]phenyl]amino]-1,3,5-

triazin-2-yl]amino]phenyl]azo]naphthalene-1,3,6-trisulphonic acid, sodium salt (CAS No. 94158 -80 -2) were reviewed to summarize the following information:

72 hrs aquatic toxicity study was predicted (SSS QSAR prediction model, 2016) to assess toxic effects of the test compound7-[[2-[(aminocarbonyl)

amino]-4-[[4-chloro-6-[[4-[vinylsulphonyl]phenyl]amino]-1,3,5-triazin-2-yl]amino]phenyl]azo]naphthalene-1,3,6-trisulphonic acid, sodium salt (CAS No. 94158 -80 -2) and the results were predicted. The study was based on the effects of the test compound on Scenedesmus subspicatus (new name: Desmodesmus subspicatus) in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the testcompound 7-[[2-[(aminocarbonyl)amino]-4-[[4-chloro-6-[[4-[vinylsulphonyl]phenyl]amino]-1,3,5-triazin-2-yl]amino]phenyl]azo]naphthalene-1,3,6-trisulphonic acid, sodium salt (CAS No. 94158 -80 -2) was estimated to be 102.527 mg/l on the basis of growth rate. Thus, based on this value, it can beconcluded that the substance can be considered as non-toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.

naphthalene-1,3,6-trisulphonic acid, sodium salt(CAS No. 94158 -80 -2) with growth inhibition effects. Thus, based on this value,it can be concluded that the substance can be considered as non-toxic to aquatic organisms and thus cannot be classified as hazardous as per the CLP criteria.

Various predicted studies and read across study for the 1,3,6-Naphthalenetrisulfonic acid, 7-(2-(2-((aminocarbonyl)amino)-4-((4-chloro-6-((4-(ethenylsulfonyl)phenyl)amino)-1,3,5-triazin-2-yl)amino)phenyl)diazenyl)-, sodium salt were reviewed to summarize the following information:

The study was based on the effects of the 1,3,6-Naphthalenetrisulfonic acid, 7-(2-(2-((aminocarbonyl)amino)-4-((4-chloro-6-((4-(ethenylsulfonyl)phenyl)amino)-1,3,5-triazin-2-yl)amino)phenyl)diazenyl)-, sodium salt on the Daphnia magna in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the 1,3,6-Naphthalenetrisulfonic acid, 7-(2-(2-((aminocarbonyl)amino)-4-((4-chloro-6-((4-(ethenylsulfonyl)phenyl)amino)-1,3,5-triazin-2-yl)amino)phenyl)diazenyl)-, sodium salt was estimated to be 65.576499939 mg/Lon the basis of mobility.

 

48 hrs aquatic toxicity study (EPI suite, 2016) was conducted to assess toxic effects of the test compound 1,3,6-Naphthalenetrisulfonic acid, 7-(2-(2-((aminocarbonyl)amino)-4-((4-chloro-6-((4-(ethenylsulfonyl)phenyl)amino)-1,3,5-triazin-2-yl)amino)phenyl)diazenyl)-, sodium salt (CAS no 94158-80-2) and the results were predicted. The study was based on the effects of the test compound on Daphnia magna in a static fresh water system. The predicted data suggests the effective concentration (EC50) for the test compound 1,3,6-Naphthalenetrisulfonic acid, 7-(2-(2-((aminocarbonyl)amino)-4-((4-chloro-6-((4-(ethenylsulfonyl)phenyl)amino)-1,3,5-triazin-2-yl)amino)phenyl)diazenyl)-, sodium salt was estimated to be 7848.481 mg/l on the basis of mobility. 

 

As in vitro experiment for read across CAS name suramin (145 -63 -1) (Teresa Santamarina, Florencio M Ubeira, José Manuel Leiro, Manuel Luis Sanmartín Durán, Bull. Eur.Ass Fish path 13(2), 45, 1993) was performed on 8 Gyrodactvlus (worm) were taken from fish and placed in petri dish with added suramin and observed for 30 and 60 min after exposing to the suramin. A control trial was also used were zero mortality observed. After exposure period only 1 worm death was observed.On the basis of this, the LD 50 value for aquatic invertebrates for suramin (145-63-1) was determined to be greater than 200 mg/l.

 

Based on the result of SSS QSAR prediction model the test chemical was classified as chronic category 3 but as the test chemical was log Kow value 0.48 which was less than 4. On the basis of this the 1,3,6-Naphthalenetrisulfonic acid, 7-(2-(2-((aminocarbonyl)amino)-4-((4-chloro-6-((4-(ethenylsulfonyl)phenyl)amino)-1,3,5-triazin-2-yl)amino)phenyl)diazenyl)-, sodium salt was considered as not classified as per QSAR prediction,EPI suite,2016, read across study and also according to the CLP regulations.
