Each test concentration, the control and the solvent control were analytically verified once before the day of introduction of the fish and daily after the start of the exposure.

DMF was used as solvent with a loading rate of 0.1 mL/L. The solvent concentration was the same in all concentration levels and the solvent control (0.100 mL/L).
Details on test solutions:
Stock solution preparation:
A stock solution of 2000 mg/L was prepared in DMF. An appropriate volume of the test item was pipetted into a glass flask and filled up with the solvent. Density was taken into account. The solution was agitated until it is visually clear dissolved.
Further stock solutions were prepared by dilution with DMF. The stock solution was prepared in appropriate intervals of maximum 7 days.
The stock solutions were diluted considering the flow rate of the system. Test solutions flew through the test vessels for 14 days prior to the start of the exposure until measured concentrations of the test item show no trend of increasing or decreasing.

- Source: All fish used in the test were gained at Noack Laboratorien GmbH from a single brood stock (supplier: Umweltbundesamt,
Schichauweg 58, D-12307 Berlin)

HOLDING
Holding was performed at the test facility at 23 ± 2 °C and diffuse light (540 – 1000 Lux, photoperiod of 12 to 16 h light / 12 to 8 h
dark). The water was changed at least once per week. The dissolved oxygen concentration was more than 80% of the air
saturation value.
Zebrafish with at least 9 days of acclimatization and mortality < 5% within these days prior to the start of the exposure were used in
the test.
No mortality was observed during this time. No disease treatments were administered throughout holding and testing.

The test substance showed a very steep concentration response curve.
The highest test concentration of 78.5 µg/L arithmetic mean measured (200µg/L nominal) resulted in 86% mortality. In the next lower concentration of 27.3 µg/L arithmetic mean measured (90.0 µg/L nominal) no mortality was observed.

Conclusions:
In this study the test substance was found to cause lethal effects to Zebrafish after 96 hours at a nominal test item concentration of 200 µg/L, corresponding to an arithmetic mean measured concentration of 78.5 µg/L. The LC50 after 96 hours was 144 mg test item/L (nominal concentration), corresponding to an arithmetic mean measured concentration of 50.5 mg/L. The LC0 and LC100 after 96 hours were 90.9 and > 200 mg test item/L (nominal test item concentration), respectively, corresponding to arithmetic mean measured concentrations of 27.3 and > 78.5 µg/L. The LC50-value after 96 hours was calculated by linear regression from the highest test concentration without mortality and the highest test concentration with 86% mortality.
Executive summary:

A valid guideline study according to OECD 203 over 96h resulted in a LC50 value of 50.5 µg/L (arithmetic mean measured concentration).


Due to the volatility/instability of the substance the test was done in a flow-throgh design and aquaria were covered with glass lids.
