For the determination of the stability of the test item under the test conditions and of the maintenance of the test item concentrations during the test period, duplicate samples from the test media of all test concentrations and the control (containing algae) were taken at the end of the test (after the 72 hours test period).
The concentrations of the test item were analysed for trifluoromethanesulphinate and trifluoroacetate in the duplicate test media samples from all test concentrations collected at the start and end of the test. PREPARATION AND APPLICATION OF TEST SOLUTION
- Method: The test medium of the highest test concentration of nominal 282 mg test item/L was prepared by mixing 141 mg test item into 500 mL test water by intense stirring for 10 minutes. Adequate volumes of this test medium were diluted with test water to prepare the test media of the other desired test concentrations. The test media were prepared just before introduction of the algae

- Source (laboratory, culture collection): The algae were supplied by the "Sammlung von Algenkulturen, Pflanzenphysiologisches Institut der Universität Göttingen", 37073 Göttingen, Germany.
- Age of inoculum (at test initiation): These cells were taken from an exponentially growing pre-culture, which was set up 3 days prior to the test start under the same conditions as in the test.
- Method of cultivation: The algae were cultivated in the laboratories of IBACON under standardised conditions according to the test guidelines.

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) :
- Determination of cell concentrations: The cell densities in the samples were determined by spectrophotometrical measurement. Therefore defined volumes of the algal suspensions from all replicates and from the blanks were sampled and were not replaced. The algal cell densities were calculated by subtracting the absorption of the blanks, from each of the measured absorption of the test media (with algae). The cell densities in a number of samples from one control replicate were counted by microscope after 72 hours of test duration. Out of this control seven defined dilutions were prepared and measured spectrophotometrically. Based on the counted cell densities and the absorption of the control sample and the dilutions a linear regression was performed for the calculation of the cell densities in all other samples measured spectrophotometrically during the test.

Reported statistics and error estimates:
Based on the calculated cell densities, the 72-hour ErC50 and the 72-hour EyC50, the corresponding EC20 and EC10 values and where possible their 95 %-confidence limits were calculated by Probit analysis.
For the determination of the 72-hour LOEC and the 72-hour NOEC, the calculated growth rates and yields at each test concentration were tested for significant differences compared to the control values by the Bonferroni-Welch t-test (yield and growth rate).
The software used to perform the statistical analysis was ToxRat Professional, Version 2.10.05, ToxRat Solutions GmbH.

Conclusions:
The influence of reaction mass of TFSK/TFAK on the growth of the freshwater green algae Pseudokirchneriella subcapitata was assessed in a static dose-response test. The 72-hour ErC50 value was calculated to be > 282 mg test item/L, and the 72-hour EyC50 was calculated to be 10.0 mg test item/L. The 72-hour NOErC was determined to be 2.69 mg test item/L and the associated 72-hour LOErC was 4.3 mg test item/L. The 72-hour NOEyC was determined to be 0.84 mg test item/L and the associated 72-hour LOEyC was 2.69 mg test item/L.
Executive summary:

In a 72 hour acute toxicity study, the cultures of Pseudorkirchneriella subcapitata were exposed to the reaction mass TFSK/TFAK at nominal concentrations of 0.84, 2.69, 8.6, 27.5, 88 and 282 mg/L under static conditions in accordance with the OECD 201 guideline.

Analytical verification showed an acceptable correlation between the nominal and the measured concentrations. Therefore, the results were based on nominal concentration.

The following results were determined:

EC50 growth rate > 282 mg/L of test item corresponding to >100 mg/L in active ingredient

NOEC Growth rate = 2.69 mg/L of test item corresponding to 0.955 mg/L active ingredient

EC10 Growth rate = 6.38 mg/L of test item corresponding to 2.26 mg/L active ingredient.

All validity criteria were met, the test is considered to be valid. In these test conditions, the reaction mass TFSK/TFAK is not harmful to algae.

Description of key information

In an algae toxicity study, the 72-hour ErC50 value was calculated to be > 100 mg/L of reaction mass. Therefore, the reaction mass is not harmful to algae. Additionally, the 72-hour ErC10 value was calculated to be 2.26 mg/L.

Additional information

A reliable key study on toxicity to algae is available for the reaction mass of potassium trifluoroacetate and potassium trifluoromethanesulphinate. In this study, the test item was an aqueous solution of the reaction mass of TFSK and TFAK because it is the form under which the substance is manufactured and used. The results were expressed in both test item and active matter (i.e. both dried constituents of the reaction mass).

The cultures of Pseudokirchneriella subcapitata were exposed for 72h to the reaction mass TFSK/TFAK at nominal concentrations of 0.84, 2.69, 8.6, 27.5, 88 and 282 mg/L under static conditions in accordance with the OECD 201 guideline and under GLP conditions. Analytical verification showed an acceptable correlation between the nominal and the measured concentrations. Therefore, the results were based on nominal concentration. The following results were determined:

EC50 growth rate > 282 mg/L of test item corresponding to >100 mg/L in active ingredient

NOEC Growth rate = 2.69 mg/L of test item corresponding to 0.955 mg/L active ingredient

EC10 Growth rate = 6.38 mg/L of test item corresponding to 2.26 mg/L active ingredient.
