The target substance hydrolytically degrades in water in a matter of seconds/minutes at all relevant pH 4, 7 and 9 forming the source substance. The source substance is a the ‘common breakdown product’ of the target substance. The source and target demonstrate ‘chemical similarity’ : since the source is the structurally-similar degradation product (i.e. similarity through (bio) transformation) from the target. The target substance hydrolytically degrades in water in a matter of seconds/minutes at all relevant pH 4, 7 and 9 forming the source substance. As a consequence they should be considered chemically similar substances due to routes of common (a)biotic degradation and/or metabolism and common or similar degradants. Executive summary:

The toxicity of a source substance to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined according to the principles of OECD TG 201 and EU Method C.3 in accordance with GLP. The study was conducted under static conditions with an initial cell density of 6839 cells/mL. A stock solution of 100 mg/L was prepared with dilution water and treated 10 minutes with ultrasound. Five concentrations were tested in a geometrical series with a dilution factor of 3.16 (nominal): 1.00, 3.16, 10.0, 31.6, 100 mg/L. Three replicates were tested for each test item concentration and six replicates for the control. The environmental conditions were within the acceptable limits. Three replicates of the highest test concentration, adjusted with 1M NaOH and HCl to pH 8.1 ± 0.2, were tested under the same test conditions as the test vessels as a 'pH control'. The concentrations of the test item were analytically verified by HPLC-DAD at the start and the end of the exposure of all test concentrations and the controls. At the beginning and end of the study the pH in the vessels was as follows: 0 mg/L (control) : pH 8.03 and 9.19 ; 1.00 mg/L : pH 7.96 and 9.42 ; 3.16 mg/L : pH 7.62 and 9.30 ; 10.0 mg/L : pH 7.24 and 9.22 ; 31.6 mg/L : pH 7.12 and 7.79 ; 100 mg/L : 4.00 and 4.01 and/or pH control at 100 mg/L : pH 8.25 and 9.05, respectively. Within the pH control, < 10% mean inhibition of growth rate and yield was seen at 100 mg/L. Within the test vessels, the test item was found to inhibit the growth of the freshwater green alga Pseudokirchneriella subcapitata after 72 hours with the following effect values (based on analytically confirmed nominal test item concentrations): The EC50-values with 95% confidence intervals for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were 69.7 (39.6 – 98.3) mg/L and 52.6 (39.0 – 78.5) mg/L, respectively. Three replicates of the highest test concentration, adjusted with 1M NaOH and HCl to pH 8.1 ± 0.2, were tested under the same test conditions as the test vessels as a 'pH control'. The concentrations of the test item were analytically verified by HPLC-DAD at the start and the end of the exposure of all test concentrations and the controls. At the beginning and end of the study the pH in the vessels was as follows: 0 mg/L (control) : pH 8.03 and 9.19 ; 1.00 mg/L : pH 7.96 and 9.42 ; 3.16 mg/L : pH 7.62 and 9.30 ; 10.0 mg/L : pH 7.24 and 9.22 ; 31.6 mg/L : pH 7.12 and 7.79 ; 100 mg/L : 4.00 and 4.01 and/or pH control at 100 mg/L : pH 8.25 and 9.05, respectively. Within the pH control, < 10% mean inhibition of growth rate and yield was seen at 100 mg/L. Within the test vessels, the test item was found to inhibit the growth of the freshwater green alga Pseudokirchneriella subcapitata after 72 hours with the following effect values (based on analytically confirmed nominal test item concentrations): The EC50-values with 95% confidence intervals for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were 69.7 (39.6 – 98.3) mg/L and 52.6 (39.0 – 78.5) mg/L, respectively. The EC10-values with 95% confidence intervals for inhibition of growth rate (ErC10) and yield (EyC10) after 72 hours were 38.6 (32.6 - 96.9) mg/L and 7.92 (< 1.00 - 19.4) mg/L, respectively. The NOEC-values for both inhibition of growth rate and yield after 72 hours were 3.16 mg/L.
For the target substance: the ErC50 is considered to be 69.7 (39.6 – 98.3) mg/L, the ErC10 is considered to be 38.6 (32.6 - 96.9) mg/L and the NOEC 3.16 mg/L based on analytically confirmed nominal concentrations.


Note: The source (benzene-1,3,5-tricarboxylic acid) and target (benzene-1,3,5-tricarbonyl trichloride) substances are related by virtue of the source substance being a ‘common breakdown product’ of the target substance. The target substance hydrolytically degrades in water in a matter of seconds/minutes at all relevant pH 4, 7 and 9 forming the source substance. The read-across hypothesis has been demonstrated experimentally in other endpoints. Refer to 'Biodegradation in water: screening tests - AE 1' section for further information.
