Acute oral toxicity:

In different studies, 1-bromo-3-methoxybenzene has been investigated for acute oral toxicity to a greater or lesser extent. Often are the studies based on in vivo experiments and estimated data in rodents, i.e. most commonly in rats for 1-bromo-3-methoxybenzene along with the study available on structurally similar read across substance Benzophenone (CAS no 119-61-9). The predicted data using the OECD QSAR toolbox has also been compared with the experimental studies.

In a prediction done by SSS (2017) using the OECD QSAR toolbox with log kow as the primary descriptor, the acute oral toxicity was estimated for1-bromo-3-methoxybenzene. The LD50 was estimated to be 3542.6 mg/kg bw when rats were orally exposed with 1-bromo-3-methoxybenzene.

In another supporting experimental study given by Opdykeet al(Food and Cosmetics Toxicology. Vol. 11, Pg. 873, 1973) on structurally similar read across substance Benzophenone (CAS no 119-61-9). rats were treated with Benzophenone in the concentration of 10000 mg/kg bw orally. No mortality was observed in treated rats at 10000 mg/kg bw. Therefore, LD50 was considered to be >10000 mg/kg bw when rat were treated with Benzophenone orally. 

Also it is further supported by experimental study given by U.S. National Library of Medicine Database (ChemIDplus A Toxnet Database, 2017) on structurally similar read across substance Benzophenone (CAS no 119-61-9). Mice were treated withBenzophenone in the concentration of 2895 mg/kg bw orally. 50 % mortality and Tremor, Somnolence (General Depressed Activity) were observed in treated mice at2895mg/kg bw.Therefore, LD50 was considered to be 2895 mg/kg bw when mice were treated with Benzophenone orally. 

Thus, based on the above studies and predictions on 1-bromo-3-methoxybenzene and its read across substances and by applying weight of evidence, it can be concluded that LD50 value is greater than 2000 mg/kg bw. Thus, comparing this value with the criteria of CLP regulation, 1-bromo-3-methoxybenzene can be “Not classified” for acute oral toxicity.

Justification for classification or non-classification

Based on weight of evidence for target 1-bromo-3-methoxybenzene (CAS no 2398-37-0) and structurally similar read across Benzophenone (CAs no 119-61-9) is likely to be non hazardous by oral route of exposure and can be concluded that the substance is considered to non toxic by oral route as per the criteria mentioned in CLP regulation.
