Various studies have been investigated for the test chemical, Aluminium, 3-hydroxy-4-[(4-sulfo-1-naphthalenyl)azo]-2,7-naphthalenedisulfonic acid complex to observe the potential for dermal sensitization to a greater or lesser extent.These studies include in vivo studies in guinea oigs along with predicted data for the target chemical and its structurally similar read across substances, copper, 5-(acetylamino)-4-hydroxy-3-[[2-hydroxy-4-[[2-(sulfooxy)ethyl]sulfonyl]phenyl]azo]-2,7-naphthalenedisulfonic acid complex(C.I. Reactive Violet 5)[CAS:12226-78-9 ] and Disodium 8-acetamido-1-hydroxy-2-phenylazo-naphtalene-3,6-disulphonate(Acid Red 1)[CAS: 3734-67-6].The experimental results are also supported by theresults obtained from OECD QSAR toolbox.

Aluminium, 3-hydroxy-4-[(4-sulfo-1-naphthalenyl)azo]-2,7-naphthalenedisulfonic acid complex was predicted to be not sensitizing to the skin of male and female Dunkin Hartley guinea pigs.

This result is supported by the experimental study performed by Bianca Maria Manzini et.al (Contact Dermatitis, 1996, 34,172-175) to determine the degree of sensitization caused by the structurally similar read across substance, 5-(acetylamino)-4-hydroxy-3-[[2-hydroxy-4-[[2-(sulfooxy)ethyl]sulfonyl]phenyl]azo]-2,7-naphthalenedisulfonic acid complex(C.I. Reactive Violet 5)[CAS:12226-78-9] on human skin. 1813 patients aged 3 to 73 years underwent patch tests in the Allergological Department, University of Modena, Italy with the GIRDCA standard series. The dye was dispersed in 5% petrolatum, chosen after preliminary studies with 4% and 10%. Patches were applied to healthy skin of the back with Finn Chambers on Scanpor tape. The patches were removed after 3 days and reactions were read 3 hours later at 4 days. Reactions were evaluated according to ICDRG Guidelines.

However it was not known whether the sensitizing components of the reactive dye molecules are the same as those responsible for occupational IgE- mediated and contact allergy, or if they are new groups derived by the end of the dying process. Also no particular association was found between sensitization to reactive dye and sensitization to standard allergens .Hence. C.I. Reactive Violet 5(12226-38-9) was considered to be not sensitizing to humans.

The above results are supported by the study summarized in Scientific Committee on Cosmetology (seventh series), 1988; for the structurally similar read across substance,Disodium 8-acetamido-1-hydroxy-2-phenylazo-naphtalene-3,6-disulphonate(Acid Red 1)[CAS: 3734-67-6].Guinea pigs were given two times 4 intradermal injections of 0.1 ml of a 7.5% solution in saline during the induction phase.14 days after second injection, a challenge exposure was conducted. In the challenge exposure, single intradermal injection of a 3% solution in saline was administered to the guinea pigs.

Signs of sensitization were not observed. Hence RED 2G (3734-67-6) was considered to be non sensitizing after challenge exposure in guinea pigs.

Based on the available data for the target as well as read across chemicals and applying the weight of evidence approach, Aluminium, 3-hydroxy-4-[(4-sulfo-1-naphthalenyl)azo]-2,7-naphthalenedisulfonic acid complexcan be considered to be not sensitizing to skin.Comparing the above annotations with the criteria of CLP regulation, test chemical can be classified under the category “Not Classified”.
