Description of key information

The 21d NOEC reproduction for daphnia of 32 µg/L determined in an OECD 211 test using river water with N-C 12-14-alkyl-1,3-diaminopropane (CAS 90640-43-0) was used as a key value for the registered substance Amines, N-C16-22-alkyltrimethylenedi.

Toxicity daphnia tests are not available for Amines, N-C16-22-alkyltrimethylenedi. Based on similarities of the environmental fate properties and the ecotoxicity data obtained in algae, Amines, N-C16-22-alkyltrimethylenedi can be grouped with the alkyl 1,3 diaminopropanes into the same category. In order to generate the worst-case scenario, all the OECD 211 tests done with various alkyl chain length diamines were taken into account and the lowest EC10 value was used as a key value for the registered substance Amines, N-C16-22-alkyltrimethylenedi.

The Daphnia magna Reproduction Test with natural river water (Semi-Static, 21 d) of N-C 12-14-alkyl-1,3-diaminopropane (CAS 90640 -43 -0) was conducted according to OECD 211 (Noack, 2008a). The test method was semi-static. All concentrations and control were analytically verified at four sampling dates with LC-MS/MS. Reproduction was the most sensitive endpoint. The effect levels (EC50, NOEC and LOEC) are therefore based on the reproduction output as the most sensitive effect. The NOECreproduction was 32.0 µg/L and the LOECreproduction was 100 µg/L determined after 21 days. The EC50 parental mortality was 179 µg/L determined after 21 days of exposure. The later value is used as worst-case acute EC50 for aquatic invertebrates in the hazard assessment in the absence of reliable acute data. The effects are expressed as nominal values because the tests were performed with river water as they are intended to be used in an evaluation of the environmental risks based on the bulk approach.

Daphnia reproduction tests have been performed in the same conditions with C18-unsatd-1,3-diaminopropane, CAS 7173 -62 -8 (Noack, 2008b) and C16 -18 and C18 -saturated-1,3 -diaminopropane, CAS 133779 -11 -0 (Noack, 2008c). For the C18 -unsatd, the nominal test item concentrations 320 and 1000 µg/L had a biologically significant effect on adult mortality (> 20 %) after 21 d of exposure and the EC50 for the adult mortality was calculated to be 290 µg/L. However, effects on reproduction did not occur.

For the C16 -18 and C18 -satd, the NOEC for adult mortality was 320 µg/L. 100% adult mortality was observed at the next higher concentration of 1000 µg/L. The EC50 for adult mortality is 566 µg/L. However, as for the C18 -unsatd, no effects on reproduction were observed up to 320 µg/L.
