Samples for analytical determinations were taken at 0 and 48 h after mixing of the test solutions from the approximate center of the test vessels. The samples were stored at -18 to - 25 °C until analyses.

Stock solution:
0.5 g test substance was mixed with 200 mL test medium and the volume was then made up to 500 mL with test medium. The stock solution was homogenized for five minutes in an ultrasonic bath.

The Daphnia were bred in the testing facility of the Product Ecology TF, Textile Dyes Division under standardised conditions.

BREEDING:
Cultures of Daphnia magna were maintained in glass vessels containing approx. 3.0 L of reconstituted water at 18-22 °C. The water was renewed partially three times weekly. The Daphnia were fed with a suspension of green algae (Scenedesmus subspicatus) supplemented by a suspension of TETRAMIN-extract in such quantities that the food was consumed within 24 h. The quality of the Daphnia was checked every 3 months by determining the EC50 values (24 h) for potassium dichromate (range: 0.8-1.5 mg/L).

24 h before the start of the exposure, reproductive daphnids were separated from the young Daphnia by sieving all individuals through a 710 µm sieve.

Executive summary:

A study was conducted to determine the acute toxicity of the test substance FAT 92368/A to Daphnia magna in a 48 h static test according to the OECD Guideline 202, Section 2, and the EU Method C.2, in GLP-compliance. Daphnia were exposed to nominal test substance concentrations of 10, 18, 32, 58, 100 and 180 mg/L. Analytical determination of the test substance concentration was carried out at 0 and 48 h by spectrophotometry. During the test, the test substance concentrations in the analysed test media were in the range of 91.0 - 100.0 % of the nominal values. No alteration of the test substance was observed after 48 h. The average measured test substance concentrations were 9.2, 18, 32, 56, 99 and 175 mg/L. No mortality was seen with blank and at 9.2 mg/L, 10 and 45 % mortality was seen at 18 and 45 mg/L, respectively, while 100 % mortality was seen with 56, 99 and 175 mg/L test concentrations. Based on the above observations, the 24 and 48 h EC0 values for the test substance were established at ≥9.2 mg/L, and the 24 and 48 h EC100 values were established at 56 and 175 mg/L respectively. The EC50 values were calculated with the ‘Probit model’ method and found to be 47.4 mg/L at 24 h and 30.5 mg/L at 48 h. Hence,based on the above findings, the test substance is considered to be slightly toxic to Daphnia with EC50 values of 47.4 and 30.5 mg/L at 24 and 48 h exposures, respectively.
