PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Chemicals:
Calcium solution, 0,5 M: 109,55 g calciumchloride-6-hydrate, CaCl2 6H20, clean crist., is solved in deionized water and completed to a volume of 1L, 1 ml of the solution contains 0,5 mmol calcium-ions.
Magnesium solution, 0,5 M: 123,25 g magnesiurnsulfate-7-hydrate, MgSO4.7 H2O, clean krist., is solved in deionized water; the solution is completed to a volume of 1 L with deionized water. 1 ml of the solution contains 0,5 mmol magnesium-ions.
Hydrogencarbonate solution, 1 M: 84,01 g potassiumhydrogencarbonate, NaHCO8, clean, is solved in deionized water; the solution is completed to a volume of 1 L with deionized water. 1 ml of the solution has an acid capacity of 4,3 (formerly known as m-value) mmol/L when completed to a volume of 1 L water.
Sulfic acid, 1 N; 1 ml of the solution has an acid capacity of 4,3 mmol/L when completed to a volume of 1 L water.
Testing water: for the study chloric free drinking water from a local source was used with a concentration of calcium-magnesium-ions of (2,7 ± 0,5) mmol/1 - corresponding to (15 ± 3) °d; the ratio of calcium- to magnesium-ions needs to be approx. 4 to 1.
The acid capacity 4,3 needs to be 0,7 to 0,9 mmol/1 - corresponding to 2,0 to 2,5 °KH - and the pH-value needs to be between 7 and 8 (this pH value is automatically adjusted during aeration of the water)
When the concentration of calcium- and/or magnesium-ions in the available water is to high (>3 mmol/l and/or >18 °d), it has to be adjusted by diluting with deionized water.
When the ion concentration is to low (<2 mmol/l and/or. >12 °d), it has to be adjusted by adding a calcium-and/or. magnesium solution.
When the acid capacity is to high, 1 N sulifc acid is added to the water in an in appropriate quantity.
When the acid capacity is to low, 1 M hydrogencarbonate solution is added to the water in an appropriate quantity.

A 48 h LC0 of 100 and 250 mg/l was determined. The 48h LC0 values were 100 and 250 mg/l, 48 h LC50 values were 160 and 325 mg/l and the 48 h LC100 values were 250 and 400 mg/l.

Although generally preference would be given to 96 h toxicity tests for short term toxicity to fish, in this case the 48h toxicity test was selected, because: a) this was the best test (best described) and b) the observed LC50 was the lowest observed value
