A stock preparation with a test item concentration of 100 mg/L was freshly prepared. Out of the stock solution 5 concentration levels were tested in a geometrical series with a dilution factor of 0.2, 0.632, 2.0, 6,32 and 20 mg/L (nominal). The concentration levels are based on the results of a preliminary range finding test.

EFFECT PARAMETERS MEASURED
- Determination of cell concentrations: The cell density was measured daily via Chlorophyll a- fluorescence, excitation at 436 nm, emission at 685 nm. Dilution water was used as a background signal.
Microscopic evaluation of the cells at the start and the end of the incubation period was carried out. The cells were checked for unusual cell shapes, colour differences, differences in chloroplast morphology, flocculation, adherence of algae to test containers and agglutination of algae cells.
The pH-values at the start of exposure were measured in one additional replicate of each test item concentration and the control. At the end of exposure, the pH-values were measured from pooled samples of each test item concentration and the control. The room temperature was measured continuously. Light intensity was measured prior to the start of exposure.

Results with reference substance (positive control):
The toxicity of potassium dichromate (SIGMA, batch number MKBV0900V, purity 99.0 %, CAS RN 7778-50-9) to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined over a period of 72 hours from 2017-10-10 to 2017-10-13. The reference item toxicity is in the valid range following test facility SOPs. The EC50 based on growth rate was 0.535 mg/L (Validity range: 0.747 ± 0.524), the EC50 based on yield was 0.320 mg/L (Validity range: 0.398 ± 0.291).
Reported statistics and error estimates:
The NOEC and LOEC were determined by calculation of statistically significant differences of growth rates and yield. As a standard, One Way Analysis of Variance (ANOVA) and Dunnett's test were used for NOEC/LOEC calculations. When running a One Way Analysis of Variance, a Normality test and an Equal Variance test were done first. P-values for both Normality and Equal Variance tests are 0.05. The α-value (acceptable probability of incorrectly concluding that there is a difference) is α=0.05.
