Data on the long-term toxicity to Daphnia magna of an analogous substance (EC 401-540-3, CIBA-GEIGY 1990a) and a long-term test of a mixture of stuctural isomers are available (NOTOX B.V. 2006).

In both cases the overall NOEC for reproduction in a long term study on daphnids was found to exceed the range of solubility. No toxic effects occur within the range of solubility.

The read-across substance (EC 401-540-3) and the mixture of structural isomers have similar physicochemical, toxicological and ecotoxicological properties. They are also of low water solubility and degradation and hydrolysis are not likely. Due to the inert structure no break-down products will occur in the environmental compartments.

The first read-across substances (EC 401-540-3) differs by a chlorine in para-position of both phenyl rings instead of a second phenyl ring. The second read-across substance is a mixture of the same molecule with chlorine in both para-position, para- and meta-position and both meta-postion instead of second phenyl rings. Therefore, the molecular diameter is slightly smaller.

The common functional group is the 2,5 -dihydro-pyrrolo(3,4 -c)pyrrole-1,4 dione core. It is the chromophore that is both poorly soluble in organic and in inorganic solvents. It can be assumed based on the chlorines that the water solubility in both cases is predicted to be slightly better and worst case substances have been tested.
