Determination of the test item
Analytical evaluation of the saturated solution of Savinyl Yellow RLS and the control was carried out via HPLC-DAD at the start (0 hours) and at the end of the exposure (72 hours) with algae.

Preparation of the saturated solution
The saturated solution (100 mg/L test item were weighed out) was prepared with demineralized water two days prior to the start of the exposure (0 h). The dispersion was stirred for 48 hours (1100 rpm, room temperature) with a magnetic stirrer. Undissolved particles were removed by membrane filtration (membrane filter 0.45 µm, RC, MACHEREY-NAGEL). The filter was saturated in order to avoid adsorption during the filtration. The first 25 mL filtrate were discarded.
The filtration was interrupted for 15 minutes to allow for adsorption and saturation of the filter material with dissolved test item. Thereafter, the filtration was continued. The next 25 mL were discarded. After the separation treatment the components of the dilution water were added to the saturated solution. The following filtrate, i.e. the saturated solution, was used in the test. During filtration the filter was always kept covered.

Test concentrations (nominal)
Five dilution levels of the saturated solution were tested in a geometrical series with a dilution factor of 3.16: 0.100 - 0.316 - 1.00 - 3.16 - 10.0 % of the saturated solution. The dilution levels are based on the results of a preliminary range finding test (non-GLP, Erlenmeyer flasks, with headspace in OECD TG 201 medium).

-The cell density was measured daily via Chlorophyll-a-fluorescence, excitation at 436 nm, emission at 685 nm. Dilution water was used as background signal.Self-fluorescence was observed in the definitive test. A separate replicate for the concentration levels were prepared without algae and treated like the other test replicates. The self-fluorescence of each measuring point was taken into account in the evaluation. The adsorption of the test item at the relevant wavelength (436 nm) was considered not relevant for the measurement of the fluorescence.

The toxicity of potassium dichromate (SIGMA-ALDRICH, batch number MKBV0900V, purity 99.0%, CAS RN 7778-50-9) to the unicellular freshwater green alga Desmodesmus subspicatus was determined over a period of 72 hours from 2017-10-09 to 2017-10-12. The reference item toxicity is in the valid range following test facility SOPS.

EC10-, EC20- and EC50-value with confidence intervals of yield inhibition after 72 hours was calculated by sigmoidal dose-response regression.

NOEC, LOEC and statistical analyses
NOEC/LOEC was determined by calculation of statistical significance of growth rates and yield. As standard One Way Analysis of Variance (ANOVA) and DUNNETT’s test was used for NOEC/LOEC calculations. When running a One Way Analysis of Variance a Normality test and an Equal Variance test were done first. P-values for both Normality and Equal Variance tests are 0.05. The -value (acceptable probability of incorrectly concluding that there is a difference) is =0.05.

Conclusions:
In this study, the test item was found to inhibit the growth rate of the freshwater green alga Desmodesmus subspicatus after 72 hours with the following values: The EC50-values with 95 % confidence intervals for inhibition of growth rate (ErC50) and yield (EyC50) after 72 hours were 0.353 (0.341 – 0.366) and 0.155 (0.114 – 0.224) mg/L, respectively. The NOEC-values for both inhibition of growth rate and yield after 72 hours were 0.113 mg/L, respectively. All effect levels are given based on the geometric mean measured test item concentration of the test item.
Executive summary:

The toxicity of the test item to the unicellular freshwater green alga Desmodesmus subspicatus was determined according to the principles of OECD 201 (2011) and Council Regulation (EC) 266/2016/Method C.3 (2016) .The aim of the study was to assess the effects on growth rate and yield over a period of 72 hours.

 

The study was conducted under static conditions with an initial cell density of 5836 cells/mL. A saturated solution with a nominal concentration of 100 mg/L was prepared in demineralized water two days prior to the start of exposure. The dispersion was stirred with approximately 1100 rpm for 48 hours at room temperature with a magnetic stirrer. Undissolved particles were removed by membrane filtration (membrane filter 0.45 µm, RC,Macherey-Nagel). The saturated solution and four dilution levels of the saturated solution were tested in a geometrical series with a dilution factor of √10:0.100 - 0.316 - 1.00 - 3.16 - 10.0% of the saturated solution, corresponding to the geometric mean measured test item concentrations 0.0750 – 0.0750 – 0.113 – 0.230 – 1.22 mg/L (the two lowest concentration lowest were < LOQ).Three replicates were tested for each test item concentration and six replicates for the control. The environmental conditionswere within the acceptable limits.

 

The test media were colored yellow in a concentration-related manner. The concentrations of the test item Savinyl Yellow RLS were determined at the start of the exposure (0 hours) and at the end of the exposure (72 hours) in all test media and the control via HPLC-DAD. The measured concentrations of the test item at the start of the exposure (0 hours) ranged from < LOQ to 2.52 mg/L. The measured concentrations in the old media (72 h) were in the range from < LOQ to 0.589 mg/L.

 

All effect values of the test item are given based on the geometric mean measured test item concentration.
