In different studies, Hydrogen [4-[4-(dimethylamino)-α-(2-hydroxy-3,6-disulphonato-1-naphthyl)benzylidene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium, monosodium salt (3087-16-9) has been investigated for potential for dermal sensitization to a greater or lesser extent. The prediction and studies are based on in vivo experiments in guinea pig for target chemical Hydrogen [4-[4-(dimethylamino)-α-(2-hydroxy-3,6-disulphonato-1-naphthyl)benzylidene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium, monosodium salt (3087-16-9)and its structurally similar read across substances Tetra sodium 4,4'-bis[[4-[bis(2-hydroxyethyl)amino]-6-(4-sulphonatoanilino)-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate](16470-24-9)andTetra sodium 4,4'-bis[[4-[bis(2-hydroxypropyl)amino]-6-[(4-sulphonatophenyl)amino]-1,3,5-triazin-2-yl]amino]-stilbene-2,2'-disulphonate(67786-25-8) . The predicted data using the OECD QSAR toolbox have also been compared with the experimental data of read across.

Further supported by experimental data conducted by OECD SIDS SIAM 13 (SIDS Initial Assessment Report for SIAM 13, 2001) on structurally similar read across substance Tetra sodium 4,4'-bis[[4-[bis(2-hydroxyethyl)amino]-6-(4-sulphonatoanilino)-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate](16470-24-9)on human.The read across substances share high similarity in structure and log kow .Therefore, it is acceptable to derive information on skin sensitization from the analogue substance. The skin sensitization study was performed by repeated insult patch test in 103 female volunteers using 0.1% aqueous solution. In induction phase, 10 repeated patch applications was done using 0.1% test solution for 48hr per application at back of the volunteers. In challenge phase, After 14 days rest period challenge application of 0.1% test solution was applied on back of the subject for 48hr.No skin sensitizing reaction observed after 48hr of challenge application .Hence it is considered thatTetra sodium 4,4'-bis[[4-[bis(2-hydroxyethyl)amino]-6-(4-sulphonatoanilino)-1,3,5-triazin-2-yl]amino]stilbene-2,2'-disulphonate](16470-24-9)was not skin sensitizing in human.

It is further supported by an experimental study conducted  (EPA) Office of Pollution Prevention and Toxic Substances (OPPTS) (U.S. Environmental Protection Agency Hazard Characterization Document, 2012)on structurally similar read across substance Tetra sodium 4,4'-bis[[4-[bis(2-hydroxypropyl)amino]-6-[(4-sulphonatophenyl)amino]-1,3,5-triazin-2-yl]amino]-stilbene-2,2'-disulphonate(67786-25-8)in guinea pig .The skin sensitization study of tTetra sodium 4,4'-bis[[4-[bis(2-hydroxypropyl)amino]-6-[(4-sulphonatophenyl)amino]-1,3,5-triazin-2-yl]amino]-stilbene-2,2'-disulphonate(67786-25-8)was performed on 14 guinea pigs, by using 0.05ml suspension of 0.1% of test material in physiological saline. In induction phase, initially 0.05ml suspension of 0.1% of Fluorescent Brightener 263, tetra sodium salt in physiological saline was given by intacutaneous injection. After that 9 additional injection of 0.1ml suspension was given.In challenge phase , after 14 days rest period of last induction injection 0.5 ml of suspension injection was given as challenge ,no sensitization observed .Hence it is considered that Fluorescent Brightener 263, tetra sodium salt (67786-25-8) was not skin sensitizing in guinea pig.

Thus based on the above predictions on Hydrogen [4-[4-(dimethylamino)-α-(2-hydroxy-3,6-disulphonato-1-naphthyl)benzylidene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium, monosodium salt (3087-16-9) as well as its read across substances and applying weight of evidence, it can be concluded that Hydrogen [4-[4-(dimethylamino)-α-(2-hydroxy-3,6-disulphonato-1-naphthyl)benzylidene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium, monosodium salt is not a skin sensitizer. Thus comparing the above annotations with the criteria of CLP regulation, Hydrogen [4-[4-(dimethylamino)-α-(2-hydroxy-3,6-disulphonato-1-naphthyl)benzylidene]cyclohexa-2,5-dien-1-ylidene]dimethylammonium, monosodium salt (3087-16-9) can be considered as not classified for skin sensitization effects.
