In a 4-week rat inhalation study performed according to OECD Guideline 412 at concentrations of 500, 1500, 5000, and 15000 ppm, no NOAEC was observed (LOAEC of 500 ppm) based on myocardial vacuolation and focal and multifocal mononuclear cell infiltrate ascribed to a direct toxic effect on the myocardial fibers and to the effects on blood vasculature, respectively (TNO, 2010).Additionally in a second 4-week rat inhalation study (Huntingdon Life Sciences, 2010). performed according to Guideline 412 at measured concentrations of 4967, 14953, and 49686 ppm, no NOEC was determined based on myocardial vacuolation/myofibre degeneration in all groups. Myocardial fibrosis and myocardial congestion (males) was observed at 14593 and 49686 ppm. Therefore, the test substance is classified as as Xn; R48/20 (Harmful: danger of serious damage to health by prolonged exposure through inhalation) according to EU Directive 67/548/EEC and STOT Rep Exp. 2 (H373: May cause damage to organs [blood vessels; heart] through prolonged or repeated inhalation exposure) according to EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008.
