No toxic effects up to the limit of water solubility (< 0.05 mg/L in aqua dest) for fish

No studies investigating the short-term toxicity of Diisooctyl adipate (CAS 1330-86-5) to freshwater fish are available. Therefore, in accordance to Regulation (EC) No. 1907/2006 Annex XI, 1.5 Grouping of substances, a read-across to the structurally related category members Dioctyl adipate (CAS 123-79-5) and Bis(2-ethylhexyl)adipate (CAS 103-23-1) was conducted. Further justification is given within the endpoint summary 6.1 and within the category justification section 13. In this case of read-across, the best suited (highest degree of structural similarity, nearest physico-chemical properties) read-across substance within this category was used.

The test with Dioctyl adipate (CAS 123-79-5) was conducted under semi-static conditions according to OECD 203 using Danio rerio as test organism (Richterich and Mühlberg, 2008). Test concentrations of 100, 3000 and 10000 mg/L were directly added to the test vessels. Mortalities were observed only at the highest test substance concentration.The LC50 (96h) were calculated as 5500 mg/L, based on nominal concentration. 

The test with Bis(2-ethylhexyl)adipate (CAS 103-23-1) was conducted under semi-static conditions according to OECD 203 using Medaka fish (Oryzias latipes) as test organism (MOE, 2000). A mixture of DMF and HCO-40 was used as vehicle at a concentration of 100 mg/L. Only one test concentration was applied (50 mg/L nominal, 45 mg/L measured). No mortalities were observed.Therefore, the LC50 (96h) is assessed as > 50 mg/L (nominal) and > 45 mg/L (measured).

The test with Diisononyl adipate (CAS 33703-08-1) was conducted under static conditions according to OECD 203 using Leuciscus idus as test organism (BASF, 1985). Test concentrations of 10, 100, and 500 mg/l were directly added to the test vessels. No mortalities were observed even at the highest test substance concentration.The LC50 (96h) was calculated as > 500 mg/L, based on nominal concentration. 

Due to structural and property similarities to the analogue substances Dioctyl adipate (CAS 123-79-5), Bis(2-ethylhexyl)adipate (CAS 103-23-1) and Diisononyl adipate (CAS 33703-08-1) these results are also applicable to read across to Diisooctyl adipate (CAS 1330-86-5).
