- Sampling method: 2.0 mL was taken at 0, 24 and 72 hours. At the end of the exposure period, the replicates with algae were pooled at each concentration before sampling.
- Sample storage conditions before analysis: None, samples were analysed on day of sampling.
- Additional information: Compliance with the quality criteria regarding maintenance of actual concentrations was checked by running a test vessel at an intermediate item concentration (WAF 10 mg/L) but without algae and samples for analysis were taken at the start, after 24 hours of exposure and at the end of the test period.

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Test solutions were individually prepared loading rates. A three-day period of magnetic stirring was applied to ensure maximum dissolution of the test item in medium. The obtained mixtures were allowed to settle for a period of one hour. Thereafter, the aqueous Water Accommodated Fractions (WAFs) were collected by means of siphoning through glass wool and used as test concentrations.

Overall, the biological results did not follow a straight dose-response curve at the three highest test concentrations. However, comparing the results of the range-finding test with the results obtained in the final test, the concentrations measured at loading rates of 100 mg/L were comparable between both tests (2.32 and 2.69 mg/L) as were the effects to some degree (11 and 27%). The reason for the stronger inhibition of algal growth at a loading rate of 32 mg/L is unknown.

Microscopic observations at the end of the test revealed a normal and healthy appearance of the algal cells exposed to 0.72 and 1.6 mg/L when compared to the control.

For determination of the NOEC and the EC50 the approaches recommended in the OECD guideline 201 were used. An effect was considered to be significant if statistical analysis of the data obtained for the test concentrations compared with those obtained in the negative control revealed significant inhibition of growth rate or inhibition of yield (Williams Multiple Sequential t-test Procedure, α=0.05, one-sided, smaller).

Additionally, the EC10 and EC20 were determined to meet the recommendations as put down in "A Review of Statistical Data Analysis and Experimental Design in OECD Aquatic Toxicology Test Guidelines" by S. Pack, August 1993.

Calculation of ECx values was based on probit analysis using linear max. likelihood regression with the percentages of growth rate inhibition and the percentages of yield inhibition versus the logarithms of the corresponding average exposure concentrations of the test item.

The calculations were performed with ToxRat Professional v. 3.2.1. (ToxRat Solutions® GmbH, Germany).

At the start of the test, the actual test concentrations increased from 0.14 to 2.7 mg/L with increasing loading rate. At the end of the exposure period, the measured concentrations had decreased to 13-36% of the initial concentrations.
