Isopropoxypropylamine is corrosive to the skin after a 3-min exposure, corrosive to the eyes and irritating to the respiratory tract.

Skin irritation/corrosion

In a key study, the acute dermal irritation of 3-isopropoxypropylamine was evaluated in one male New Zealand White rabbit according to OECD N° 404 Guideline (Manciaux, 1998a). 0,5mL of 3-Isopropoxypropylamine was applied during 3 minutes, 1 hour and 4, hours in the rabbit on clipped area. The cutaneous reaction was observed one hour and then daily to day 15 after removal of the dressing. After application of the substance during 3 minutes and 1 hour, a severe erythema (grade 4) was observed from day1 to 11. It was masked by an important necrosis of the skin thereafter. A severe oedema (grade4) was noted on day1. Necrosis and thickening of the skin were recorded from day 1or 2, up to the end of the observation period. After a 4-hour exposure, a severe erythema was observed from days 1 to 4. It was masked by an important necrosis of the skin thereafter. Necrosis and thickening of the skin were recorded from day 1, up to the end of the observation period. Under these experimental conditions, 3-Isopropoxypropylamine is considered as corrosive.

 

In a supporting study, two animals were treated for 1, 5, 15 min and 20 hours using occlusive conditions (BASF, 1970c). An application site of 2.5x2.5 cm was covered with3-isopropoxypropylamine. After the application time (1, 5, 15 min) the skin was washed with Lutrol (50%). The animals were observed for 8 days and skin changes were recorded daily. The report describes findings after 24 hours and at the end of the observation period (8 days). For a final evaluation, the findings after 48 and 72 hours from the raw data were taken into account. The application of 3-isopropoxypropylamine caused necrosis to the exposed tissue and therefore a precise evaluation of the erythema and edema scores was not possible. On the day of application marked erythema was noted. 24 h post application leathery to hard black-reddish necrosis was noted for all exposure intervals.

 

Eye irritation/corrosion

Fifty µL of 3-isopropoxypropylamine were applied to the conjunctival sac of one eye in 2 animals (BASF, 1970d). The animals were observed after 10 min, 1 and 3h on the day of treatment and up to 8 days afterwards. Findings were recorded daily. The eyes were not washed out after 24 hours. The report describes findings after 1 and 24 hours and at the end of the observation period. For final evaluation, the findings after 48 and 72 hours from the raw data were taken into account. The application of 3-isopropoxypropylamine caused corrosion to the exposed eyes and therefore a precise evaluation of opacity, erythema, chemosis and iritis was not possible.

Respiratory tract irritation

Irritation of the mucous membranes and corrosion of eyes and noses were observed in the acute inhalation toxicity study (BASF, 1970b).
