- Sampling method: For the analysis of the test substance concentrations, a sample was taken from freshly prepared stock solution (nominal 100 mg/L) and dublicate samples were taken from the freshly prepared test media of the nominal test concentrations 0.32, 1.0 and 3.2 mg/L and the control at the first treatment period (day 0), at a treatment period in the second week (day 12) and at a treatment period in the last week (day 16). For the determination of the stability of the test substance under the test conditions, additional samples were taken at the end of two treatment periods (on day 14 and 19). All samples samples were taken out of the actual test (including the algae food).

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: The test substance was homogenized by intesive stirring. Then an adequate amount of the homogenized test substance was weighted into test water for the preparation of a stock solution (100 mg/L). The test concentrations were added to the test water without any further pretreatment. The test media were intensively stirred before introduction of daphnia.

Executive summary:

The influence of the test substance on the reproduction and survival rate of Daphnia magna was investigated in a 22-day semi-static test following the proposal for an update of OECD guideline 202, part II, draft 6/5/91. The test was performed in compliance with the GLP.

The nominal concentrations tested were 0.10, 0.32, 1.0, 3.2 and 10.0 mg/L and a control.

Analytical results:

The analytically determined test substance concentrations in the freshly prepared test media varied in the range from 75 to 108 % of nominal, in the mean over all measurements from 92 to 102 % of nominal.

During the test medium renewal periods of 48 to 72 hours, the test substance concentrations strongly decreased. In the samples, taken out of the test media of the biological test (including algal food) after 48 hours respectively 72 hours exposure periods, concentrations at or below the determination limit of the analytical method were determined. The arithmetic mean values between the measured test substance concentrations in the freshly prepared test media and the measured concentrations in the 48 hours respectively 72 hours old test media amounted to 0.18 mg/L (= 55 % of nominal) at nominally 0.32 mg/L, to 0.6 mg/L (61 % of nominal) at nominally 1.0 mg/L, and to 1.9 mg/L (55 % of nominal) at nominally 3.2 mg/L.

Biological test results:

Based on the survival rates and the reproduction rates of the test animals, the highest concentration tested without toxic effects (NOEC) after the exposure period of 22 days was determined to be 1.0 mg/L (nominal; mean measured: 0.6 mg/L). The lowest concentration tested with toxic effects (LOEC) was nominally 3.2 mg/L (mean measured: 1.9 mg/L) due to the high mortality rate of Daphnia magna at this test concentration.
