Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
- Concentrations: 0.22 mg/L
- Sampling method: (at the beginning of exposure) 0.75 ml of test solution from the middle layer of one vessels (diluted suitably with purified water if it was predicted that the concentrations will exceed the ones of the calibration curve). Next, 0.75 ml of solution and 0.75 ml of acetonitrile were mixed and the resulting solution was analysed by LC/MS.
(at the end exposure): 0.75 ml of test solution from the middle layer of one vessels (diluted suitably with purified water if it was predicted that the concentrations will exceed the ones of the calibration curve). Next, 0.75 ml of solution and 0.75 ml of acetonitrile were mixed and the resulting solution was analysed by LC/MS.
- Sample storage conditions before analysis: not indicated

Test solutions

Vehicle:
yes
Details on test solutions:
The test solutions were prepared as follows:

(1) Preparation of stock solution I: 44 mg of test substance were dissolved in 20 ml N,N-dimethylformamide and mixed by inversion. The resulting concentration was of 2200 mg/L. The test solution was fresh prepared, just before the exposure.

(2) Preparation of stock solution II: 100 ul of stock solution I were mixed with 1000 ml of dilution water. The stock solution was added to the medium while stirring, and then the resulting solution was sonicated for 30 min. The resulting concentration of stock solution II was 0.22 mg/L.

(3) Preparation of the solvent stock solution: 50 ul dimethylformamide was mixed with the 500 ml of dilution water. The stock solution was mixed by inversion.

The stock solution (2) and the solvent stock solution (3) were used as such for the final test. In both cases, the final solvent concentration was 100 ul/L. Dilution water only was used in the control group.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
Under the conditions of the test, SX-1 did not induce immobility in Daphnia up to its solubility limit (EC50 > 0.22 mg/L). Based on the current results and owing to the low water solubility (< 0.1 mg/L), SX-1 should not be classified as Acute 1.
