Study design
- Test temperature:

HPLC method
- Details on study design: HPLC method:
- EQUIPMENT
- Apparatus: Liquid chromatograph
- Type: Shimadzu LC-2010AHT with UV/VIS detector
- Type, material and dimension of analytical (guard) column: ZORBAX CN, 5 micrometrs, 150 x 4.6 mm
MOBILE PHASES
- Type: 550 mL 0.01 M citrate-buffer pH=6.1 was mixed with 450 mL of methanol
- Experiments with additives carried out on separate columns: no
- pH: 6,0
- Solutes for dissolving test and reference substances: methanol
DETERMINATION OF DEAD TIME
- Method: by inert substances which are not retained by the column
REFERENCE SUBSTANCES
- Identity: acetanilide, aniline, atrazin, benzamide, 2,5-dichloraniline, phenyl benzoate, 2-nitrobenzamide
DETERMINATION OF RETENTION TIMES
- Quantity of test substance introduced in the column: 20 micrometrs
- Quantity of reference substances: 10 micrometrs
REPETITIONS
- Number of determinations: 4 times
EVALUATION
- Calculation of capacity factors k: k=(retention time of test compound/ dead time) - 1
- Determination of the log Koc value: log Koc = (log capacity factor - intercept of calibration line)/ slope of the calibration line
Batch equilibrium or other method
- Details on test conditions:
- Deviations
The HPLC method is only applicable to chemical substances with log Koc values ranging from 1.5 to 5.0. The test substance does not meet this
requirement, at all. This leads to the retention time of the test substance much lower than that of any reference compound. A linear extrapolation is
only applicable instead of an interpolation.
It can be concluded with an assurance, that a value of the log Koc of test substance is lower than the lowest value used for calibration, however the
resulting value should be only considered as an informative one.
