No experimental data on the skin sensitisation potential of target substance was found in literature, for this reason, data on similar substances was considered together with the estimation of the OECD QSAR.


 


A structural analysis carried out on the target substance by OECD QSAR Toolbox identified some alert for protein binding for a “aromatic carbonyl compound”. However, this categorization does not apply to the target substance as its reactivity is not related to an aromatic carbonyl structure being the substance characterized by fused aromatic rings. 


Experimental data on 1,4-dihydroxyanthraquinone and 1,4,5,8-tetraaminoanthraquinone are also available but gave discordant results. 1,4-dihydroxyanthraquinone is not classified as skin sensitizer while 1,4,5,8-tetraaminoanthraquinone has a harmonized classification as skin sens. 1. The target substance could behave likewise an amino derivative in case of a systemic exposure where the endogenous enzymes could reduce the nitro group to an amino one. However, this reaction is not expected to occur in the layer of the skin responsible for an allergic response, the classification of the tetra-amino derivative is therefore not representative of the behaviour of the target substance.

Justification for classification or non-classification

Based on results provided by QSAR tools and available data on structural analogues, the substance is not classified as skin sensitising within the CLP Regulation (EC 1272/2008).
