Test solutions

Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: Due to an expected low water solubility of the test item in combination with the composition of the test item as indicated in the Technical Document, Water Accommodated Fractions were prepared individually. The preparation of test solutions started with loading rates of 1.0 to 114 mg/L (combined limit/range-finding test: 1.1 to 114 mg/L, final test: 1.0 to 100 mg/L) applying an overnight period of magnetic stirring at room temperature to ensure maximum dissolution of the test item in test medium. Subsequently, a settling period of approximately one hour followed to allow proper separation of the different liquid phases. This resulted in clear and colourless dispersions that contained undissolved floating material. Thereafter, the aqueous Water Accommodated Fractions (WAFs) were collected by means of siphoning and used as test concentrations. Siphoning was used as it is more efficient for separating liquid phases compared to filtration.
- Evidence of undissolved material (e.g. precipitate, surface film, etc): All final test solutions were clear and colourless.

- EC50/LC50: 48-h EC50 = 0.48 mg/L. The sensitivity of the daphnids was within the range determined with the historical data collected at Charles River Laboratorie Den Bosch.
Reported statistics and error estimates:
The 24h-EC50-value was calculated from the probits of the percentages of affected daphnids and the logarithms of the corresponding test item concentrations (analytically confirmed nominal concentration) using the maximum likelihood estimation method.
The 48h-EC50-value was calculated using the Trimmed Spearman-Kaerber interpolation.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
The acute toxicity of T001589 to Daphnia magna was determined in a 48 hour static test according to OECD guideline 202. Based on average exposure concentrations, the 48-h EC50 was determined to be 14 mg/L (95% confidence interval between 13 and 15 mg/L). The results of the test can be considered reliable without restriction.
