Additional information

An enhanced combined repeat-dose and reproductive and developmental effects study was performed on TPP, a related phosphite to 2 -EHDPP.  This study was conducted in rats via oral gavage and included evaluations of systemic toxicity and neurotoxicity in both the F0 and F1 generations in addition to evaluations of reproductive performance and developmental effect. 

 

Dose levels of TPP in rats were 0, 5, 15, and 40 mg/kg/day. The NOAEL for both generations for systemic effects was 15 mg/kg. There were no reproductive effects up to 40 mg/kg in the F0 generation; however significant systemic effects were observed in both generations at 40 mg/kg. In the F0 generation effects at the top dose (40 mg/kg/day) included reduced body weights, ataxia, and foot splay. In the F1 generation, toxicity was observed postnatally during lactation at 40 mg/kg/day and included increased mortality, especially on PND 0-4, and reduced pup body weights/litter PND 7-21.
