Partition coefficient (log Pow) -2 at 25 °C

The partition coefficient of the test item was evaluated in an experimental study according to the OECD Guideline 107 (1981) and the EU Method A.8 (1992). A preliminary test was first performed to estimate the partition coefficient using solubilities of the test item in pure solvents. 1 g of test item was virtuously mixed with 10 g of n-octanol for 10 minutes in an ultrasonic bath and was centrifuged for 15 minutes at 3000 r.p.m. then filtered through a 5 µm membranous filter. A sample preparation was formed using 19.2 mg of test item and 100 ml of double distilled water (192 µg/ml), from which 5 standard solutions were made by further dilution (19.2 to 134 µg/ml). The standards were quantified using a spectrophotometer at 630 nm with a 1 cm optical path, against a blank solution of n-octanol. The absorbance for each concentration was plotted graphically and the equation used to determine the log Pow value of the test item. The results obtained by the preliminary partitioning experiment demonstrated that the expected log Pow lies outside the range accessible by the flask shaking method.
