PREPARATION AND APPLICATION OF TEST SOLUTION
Dispersant: HCO-40
The provided samples and HCO-40 at 20 times its amount were dissolved in acetone, and after distilling the acetone, ion-exchange water was added, so that a stock solution with a test substance concentration of 1000 mg/L was prepared. The operation was carried out under a light-shielded condition.

Reason for selection: Since the sensitivity is similar to that of carp and it is easily obtained as a fish for testing.

Cumulative culture condition: Upon receipt, abnormal fish were removed, and the remaining fish were treated with a medication in a cumulative culture tank and held for 12 days in a flow-water state.
Domestication condition: After the cumulative culture, the fish were transferred to a domestication tank, treated with a medication, and domesticated. During the domestication, abnormal fish were removed, and the remaining fish were held for 15 days in a flow-water state at a water temperature of 25 ± 2ºC. Then, the fish were sorted, treated with a drug, and held for 38 days under flow-through conditions.

The concentration of the dispersant (HCO-40) used at that time was 5000 mg/L and the 96-h LC50 value of the dispersant was 50000 mg/L, the test at a concentration higher than that was not carried out in consideration of the influence of the toxicity of the dispersant.
