Results and discussion

Effect concentrations
Key result
Duration:
48 h
Dose descriptor:
EC50
Effect conc.:
ca. 1.97 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mobility
Details on results:
EC50 were calculated using a logistic regression analysis with a bootstrap estimation of the 95% intervals.
EC50 24h = 2.72 mg/L (2.13 – 2.76 mg/L) (nominal concentration)
EC50 48h = 1.97 mg/L (1.81 – 2.09 mg/L) (nominal concentration), corresponding to an exposure to a mixture of 0.0132 mg/L dissolved copper and 0.45 mg/L guanidine.
Results with reference substance (positive control):
Sensitivity of the test organism was checked using potassium dichromate. The EC50 24 hours was calculated using a logistic regression analysis with a bootstrap estimation of the 95% intervals.
EC50 24h: 1.12 mg/L (1.08 – 1.17 mg/L).
The EC50 24h is in the recommended range: 0.6 – 2.1 mg/L. This result confirms the appropriate sensitivity of the strain of Daphnia magna.

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
The analyses performed have shown that dissolved copper concentrations, even if they were low, were in a range that may affect the mobility of Daphnia magna. It was therefore impossible to identify which compound of test item MNP-363 was responsible for the observed effects.
Consequently, EC50 value have been expressed in concentrations of the mixture of copper (dissolved) and guanidine:
EC50 48h = 1.97 mg/L (1.81 – 2.09 mg/L) (nominal concentration), corresponding to an exposure to a mixture of 0.0132 mg/L dissolved copper and 0.45 mg/L guanidine.
