MAIN STUDY
ANIMAL ASSIGNMENT AND TREATMENT

- Criteria used to consider a positive response: a chemical is classified as a sensitizer if a threefold or greater elevation in 3H-TdR incorporation relative to vehicle control could be recorded with at least one test concentration

TREATMENT PREPARATION AND ADMINISTRATION:
- day 1: the weight of each animal was individually identified and recorded. Open application of 25 µl of the appropriate dilution of the test item, the vehicle alone or the positive control were administrated to the dorsum of each ear
- day 2 and 3: the application procedure carried out on day 1 was repeated
- day 4 and 5: no treatment
- day 6: the weight of each animal was recorded. All test and control animals were injected with 250 µl of phophte-buffered saline (PBS) containing 20 µCi (7.4e + 5 Bq) of [3H]-methyl thymidine via the tail vein. Five ours later, the animals were killed. The draining auricular lymph nodes from each ear were excised and pooled in PBS for each animal. A single cell suspension of lymph node cells (LNC) bilaterally from individual animals was prepared by gentle mechanical disaggregation through 200 µm-mesh stainless steel gauze. Lymph node cells were washed twice with an excess of PBS and precipitated with 5 % trichloracetic acid (TCA) at 4 °C for 18 h. Pellets were resuspended in 1 ml TCA and transferred to scintillation vials containing 5 ml of scintillation fluid for 3H-counting. Incorporation of 3H-methyl thymidine was measured by ß-scintillation counting as disintegrations per minute (dpm)
