Daphnia breeding was made in a thermostatically controlled room at 22°C ± 0.5, photoperiod: 16 hours of iight and 8 hours of night. For reproduction of these crustaceans Cladoceres, three bins are
used (L = 30 cm, I = 16 cm, H = 20 cm). Everyone receives 4 liters of Volvic brand water and 100 daphnids of less than 72 hours. A two-day lag in getting started each tray allows to spread the production. This one started from 5 to 6 days after seeding. The first three generations were eliminated each time (Standard AFNOR T 90-301, 1974).
The bins were filtered every 3 days. When daphnids are in their optimal phase of reproduction (8 to 10 daphnia produced per mother every 2 to 3 days), the filtration is done through 3 screens whose
meshes are respectively 1600 μm, 800 μm and 560 μm. The first retains the good breeders, the second those which must be eliminated, and the third collects the daphnia less than 72 hours usable for testing. The bins
are renewed every week.

The methodology used for carrying out the tests follows that recommended for preliminary tests of standards AFNOR T 90-301 (1974) and T 90-303 (1978)

The were carried out using a series of15 x 160 mm glass tubes corresponding to the different tested concentrations. Each tube received five daphnias less than 72 hours for a final volume of solution of 10 ml. The tubes are then plugged with plugs and placed in the dark in a thermostatically controlled room at 22°c ± 0.5. At different reading times, the mobility of crustaceans was observed and reported as well as the different manifestations of their locomotor behaviors. The concentrations are defined which prevent the daphnids from moving normally within fifteen seconds after a slight stirring of the tubes (Cl). The Authors considered as immobilized, the daphnies that only moved their antennas or which turned on themselves (phenomena observed with poorly soluble products like endosulfan, the endine ...).
