For the assessment of the aquatic toxicity short term toxicity data with fish, daphnia, algae and microorganisms are available.

Short term toxicity to fish

In a 96 h acute toxicity study according to OECD Guideline 203 rainbow trout (Oncorhynchus mykiss) were exposed to the substance at nominal concentrations of 6, 9, 12, 18 mg/L test item (corresponding to 3, 4.5, 6 and 9 mg a.i./L) using a static system. A LC0 value of 6 mg/L, LC50 of 7.5 mg/L and a LC100 of 9 mg/L were determined. The values relate to the active ingredient.

Short term toxicity to aquatic invertebrates

The test item was tested in the Acute Daphnia Toxicity Test according to the OECD Guideline No. 202 using Daphnia magna (Straus) as test organism. The experiment was carried out as a static test with the following test item concentrations: 0.5, 1, 2, 4 and 8 mg/L medium. Twenty-five daphnia were exposed to each concentration and the control. The test item induced immobilisation to the exposed Daphnia within 48 hours. The EC50 value was 2.1 mg/L, the NOEC was 1.0 mg/L.

The result is supported by an Acute Daphnia Toxicity Study with the test item were the EC50 value was 2.2 mg/L, the NOEC was 0.9 mg/L calculated both for the active ingredient.

Toxicity to aquatic algae and cyanobacteria

The test item was tested in the Alga, Growth Inhibition Test using the single cell green algae Desmodesmus subspicatus as test organism. The experiment was carried out as a static test. Based on a pretest following test substance concentrations were chosen: 0.025, 0.075, 0.225, 0.675 and 2.025 mg/L culture medium. After 0, 24, 48 and 72 hour exposure period the extinction of an aliquot of the cultures was measured photometrically for determination of algae growth. For the growth rate the EC50 value was 0.918 mg/L and the NOEC value was 0.225 mg/L after 72 h of exposure, based on nominal concentrations. The results of this study show that the test substance induced inhibition of algal growth after 72 hours of exposure.

Toxicity to microorganisms

No significant inhibition in respiration rate of the sludge was recorded at 50 mg active ingredient per litre. The duplicate measurement confirmed the result of the first measurement. The respiration rates of the controls were within 15% of each other.

The EC50 of the reference substance, 3,5-dichlorophenol, was 10 mg/L. Therefore, the test was considered to be valid.

In conclusion, under the conditions of this present test, the substance was not toxic to waste water (activated sludge) bacteria at a concentration of 50 mg/L (based on active ingredients).

The most sensitive organism to the substance was the algae Desmodesmus subspicatus with a 72 h EC50 value of 0.918 mg/L. This value is used for the calculation of the PNECs.
