Test guideline
Qualifier:
according to guideline
Guideline:
OECD Guideline 301 F (Ready Biodegradability: Manometric Respirometry Test)
Version / remarks:
1992
Deviations:
no
Remarks:
The test material rapidly hydrolyses in aqueous media and some of the hydrolysis products may be volatile, so the Manometric Respirometry Test was considered best.
GLP compliance:
yes (incl. QA statement)

Study design

Oxygen conditions:
aerobic
Inoculum or test system:
sewage, predominantly domestic, non-adapted
Details on inoculum:
A mixed population of sewage sludge micro-organisms was obtained from the final effluent stage of the Severn Trent Water PLC sewage treatment plant, which treats predominantly domestic sewage. The sample of effluent was filtered through course filter paper and maintained on aeration in a temperature controlled room at 21 °C.
Duration of test (contact time):
28 d
Initial test substance concentration
Initial conc.:
100 mg/L
Based on:
test mat.
Parameter followed for biodegradation estimation
Parameter followed for biodegradation estimation:
O2 consumption
Details on study design:
As biodegradation progresses, the micro-organisms convert oxygen to carbon dioxide which is absorobed into ethanolamine, causing a net reduction in gas pressure with in the sample flask. The pressure reduction triggers an electrolytic process, generating oxygen and restoring the pressure in the sample flask. The magnitude and duration of the current is proportional to the amount of oxygen supplied to the microorganisms. The data on the oxygen is recorded on computer.
