Guideline:
EU Method C.2 (Acute Toxicity for Daphnia)
Version / remarks:
2008
Deviations:
yes
Remarks:
appearance of test item preparations not documented at 0 h; test temperature recorded at start of test was 23 ˚C rather than 18-22˚C; positive control study overdue for twice per year rate; deviations did not affect integrity or validity of the study.

Specific details on test material used for the study:
The test substance is listed as DIPIP (CAS # 16898-52-5). The test substance was stored at room temperature in the dark.
Analytical monitoring:
yes
Details on sampling:
T
Vehicle:
not specified
Details on test solutions:
A nominal amount of test item (200 mg) was dissolved in test water with the aid of ultrasonication for 10 minutes and the volume adjusted to 2 liters to give the 100 mg/L test concentration. The pH of the 100 mg/L test concentration was adjusted to 8.0 prior to a series of dilutions being made to give further test concentrations of 10, 18, 32 and 56 mg/L.
Test organisms (species):
Daphnia magna
Details on test organisms:
The test was carried out using first instar Daphnia magna derived from in-house laboratory cultures.
Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
48 h
Test temperature:
Temperature of 21 °C to 23 °C
pH:
pH of water 7.7 to 8.1
Dissolved oxygen:
Dissolved oxygen 8.4 to 8.7 mg O2/L
Nominal and measured concentrations:
Daphnids were exposed to an aqueous solution of the test item at nominal concentrations of 10, 18, 32, 56 and 100 mg/L.
Details on test conditions:
Following preliminary range-finding tests, twenty daphnids (4 replicates of 5 animals) were exposed to an aqueous solution of the test item at nominal concentrations of 10, 18, 32, 56 and 100 mg/L for 48 hours at a temperature of 21 °C to 23 °C under static test conditions. The number of immobilized daphnia were recorded after 24 and 48 hours.
Reference substance (positive control):
yes
Remarks:
A positive control (Envigo Study Number HP54JV) used potassium dichromate as the reference item at concentrations of 0.32, 0.56, 1.0, 1.8 and 3.2 mg/L.

Validity criteria fulfilled:
yes
Remarks:
The test was considered valid since none of the control daphnids showed immobilization or other signs of disease or stress and the oxygen concentration at the end of the test was equal to or greater than 3 mg/L in the control and test vessels.
Conclusions:
The test substance did induce acute immobilisation of Daphnia magna at nominal concentrations of 32 mg/L and 56 mg/L after 48 h of exposure. The 48-h NOEC was 18 mg/L, the 48-h LOEC was 32 mg/L and the 48-hr EC50 was 28 mg/L.

Description of key information

The key study was a static acute invertebrate toxicity test on the effects of the substance on the freshwater invertebrate Daphnia magna following OECD Guideline 202. The test substance induced acute immobilisation of Daphnia magna at nominal concentrations of32 mg/L and 56 mg/Lafter 48 h of exposure. The 48-h NOEC was 18 mg/L, the 48-h LOEC was 32 mg/L and the 48-hr EC50 was 28 mg/L.
