In Acute oral toxicity,LD50 value for target substance 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7) was considered to be 4100 mg/kg bw in rats and 6200 mg/kg bw in mouse,and for different studies available on functionally similar read across substance was considered to be 6000 mg/kg bw in rats and 4360 mg/kg bw in mouse. All these studies concluded that the LD50 value is greater than 2000 mg/kg bw. Thus, comparing this value with the criteria of CLP regulation, 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7)cannot be classified for acute oral toxicity.

Acute Oral Toxicity: 

In different studies, 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7) has been investigated for acute oral toxicity to a greater or lesser extent. Often the studies are based on in vivo experimental data in rodents, i.e. most commonly in rats for 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7) along with the study available on the functionally similar read across substance. The studies are summarized as below –

The study mentioned in Danish (Q)SAR Database (2018) for the target chemical 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7) was for acute oral toxicity. Based on the QSAR prediction done using the Danish (Q)SAR Database, the LD50 was estimated to be 4100 mg/kg bw on rat for 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7) having Reliability Index: 0.56 (moderate prediction quality).

Acute oral toxicity study was performed in rats using test chemical.50% mortality was observed at dose 6000 mg/kg bw. The above studies were further supported with the another functionally similar read across substance. Acute oral toxicity study was performed in mouse using test chemical.50% mortality was observed at dose 4360 mg/kg bw. Hence,LD50 value was considered to be 4360 mg/kg bw,when mouse were treated with test chemical orally.

Thus, based on the above summarised studies on 2,3-bis[(4-methylbenzoyl)oxy]succinic acid (32634-68-7) and it’s functionally similar read across substance, it can be concluded that LD50 value is >2000 mg/kg bw.
