Description of key information

Short-term toxicity to fish:

The toxicity of Isoamyl butyrate to fish was predicted from the read-across substance Isoamyl isovalerate (CAS no. 659 -70 -1) (see also the read-across justification).

Both substances have a similar chemical structure and corresponding physicochemical properties. In addition they show similarity in their environmental fate such as biodegradability and their ecotoxic effects on aquatic organisms. Isoamyl isovalerate is therefore proposed suitable to predict the short-term toxicity to fish of the target substance Isoamyl butyrate.

Key study: The toxicity of the read-across substance (Isoamyl isovalerate) to fish was determined in a reliable GLP study according to OECD 203. The LC50 of 3.47 mg/L was estimated based on geometric mean measured concentrations. The recalculated value to correct differences in molecular weight between source (MW = 172.270 g/mol) and target (MW = 158.240 g/mol) was 3.19 mg/L.

Short-term toxicity to aquatic invertebrates:

The toxicity of the test substance to Daphnia magna was determined in a reliable GLP study according to OECD 202. After 48 hours exposure, an EC50 value of 8.12 mg/L (geometric mean measured) was estimated.

Toxicity to aquatic algae and cyanobacteria:

The toxicity of the test substance to the green freshwater algae Pseudokirchneriella subcapitata was determined in a reliable GLP study according to OECD 201. The effect values determined were the growth rate inhibition (ErCx) and the inhibition of yield (EyCx) after 72 hours. The respective values were ErC50 = 4.68 mg/L, ErC10 = 3.13 mg/L, EyC50 = 3.84 mg/L and EyC10 = 1.23 mg/L (based on geometric mean measured concentrations).

Toxicity to microorganisms:

The biodegradation of Isoamyl butyrate was determined in a reliable biodegradation test (OECD 310) including a toxicity control. No inhibitory effects were observed on microorganisms at a substance concentrations of 15.1 mg/L. The activated sludge test was, therefore, waived.
