Justification:

Classification of 2-[2-[4-(diethylamino)phenyl]vinyl]-1,3,3-trimethyl-3H-indolium acetate (CAS no. 64346-30-1) for effects in the environment:

The test chemical 2-[2-[4-(diethylamino)phenyl]vinyl]-1,3,3-trimethyl-3H-indolium acetate (CAS no. 64346-30-1)has both outdoor and indoor uses, such as in plastic construction and building materials furniture’s and toys.The test chemical herein critically and independently evaluated for the PBT.

The aim was to assess whether the PBT criterion within Annex XIII was fulfilled for2-[2-[4-(diethylamino)phenyl]vinyl]-1,3,3-trimethyl-3H-indolium acetate. The PBT criterion was herein assessed based on experimental data in conjunction with standardized environmental fate models.

 

1.Persistence assessment

 

The tested substance does meet the P criterion within Annex XIII based on the assessment that here follows:

 

1.1.Overall degradability

 

All the biodegradable information available for the target chemical 2-[2-[4-(diethylamino)phenyl]vinyl]-1,3,3-trimethyl-3H-indolium acetate(CAS no. 64346-30-1), experimental results, for the exposure period of greater than 28 days. Support that test chemical is not readily biodegradable in the environmental conditions.

In conclusion, the chemical was estimated to be persistent in water, because of its inherently biodegradable nature.

2.        Bioaccumulation assessment

 

The tested substance doesn’t meet the B criterion within Annex XIII based on the assessment that here follows:

I.                Partition Coefficient n-octanol/water of test chemical was 3.45 which was predicted by KOWWIN, indicating that the test chemical is hydrophilic in nature. The obtained log Kow is lower than threshold value 4.5, indicating chemical does not have tendency to accumulate in aquatic organisms. 

J.               Bioaccumulation: aquatic / sediment:

Various predicted data for the test chemical and supporting weight of evidence studies for its structurally similar read across substance were reviewed for the bioaccumulation end point. On the basis of overall results, BCF value is ranging from 3.162 –9.1 L/kg, which does not exceed the bioconcentration threshold of 2000, indicating that the chemical is expected to non-bio accumulative in the food chain.

From the Annex XIII of section 1.12 for PBT classification, it cannot be classified as the bio-accumulative chemical

3.    Toxicity assessment

 

The tested substance doesn’t  meet the T criterion within Annex XIII based on the assessment that here follows:

 

3.1.Mammals

 

The substance didn’t fulfil the criteria for the mammalian toxicity, neither CMR nor STOT RE as per ANNEX XIII of PBT classification.

 

3.2. Freshwater organisms

Adverse effects of the test chemical were investigated on various aquatic vertebrates, invertebrates and algae. All the tests were performed in accordance with the OECD guidelines. The Acute toxicity values of the test chemical for the various species was ranging from 0.13 mg/L to 1.086 mg/L, which classifies the chemical into aquatic chronic 1 category. Thus, the chemical is expected to be highly hazardous in nature. However, by speculations, the test results of long-term studies, are expected to have NOEC values of 0.01 mg/L. On the basis of results obtained, it can be classified as the toxic chemical in the aquatic environment and can be classified into Toxicity category as per ANNEX XIII.

 

Overall conclusion about Persistence, Bioaccumulation and Toxicity (PBT).

Based on critical, independent and collective evaluation of information summarized herein, the tested compound didn’t meet B criteria. Thus, the tested compound cannot not been classified as a PBT chemical as per Annex XIII.
