Description of key information

Prediction done using the OECD QSAR toolbox version 3.4 with log kow as the primary descriptor and considering the five closest read across substances, gene mutation was predicted for 3,7-dimethylocta-2,6-dien-1-yl propionate (105-90-8). The study assumed the use of Salmonella typhimurium strains TA 1535, TA 1537, TA 98, TA 100 and TA 102 with and without S9 metabolic activation system. 3,7-dimethylocta-2,6-dien-1-yl propionate was predicted to not induce gene mutation in Salmonella typhimurium strains TA 1535, TA 1537, TA 98, TA 100 and TA 102 in the presence and absence of S9 metabolic activation system and hence, according to the prediction made, it is not likely to classify as a gene mutant in vitro. Based on the predicted result it can be concluded that the substance is considered to not toxic as per the criteria mentioned in CLP regulation.

The study assumed the use of Salmonella typhimurium bacteria in the Ames test. The end point for gene mutation has been modeled in the Danish QSAR using the three software systems Leadscope, CASE Ultra and SciQSAR. (Environmental Mutagenesis, 1986)to determine the mutagenic nature of Geranyl acetate(105-87-3) . Geranyl acetate was examined for its ability to cause mutagenic changes when tested in strains of the bacteria Salmonella typhimurium, specifically, TA 1535, TA 1537, TA 98 and TA 100 through the Preincubation assay method. Preliminary dose range finding study was performed initially to set the doses for the main study. The test was conducted both in the presence and absence of metabolic activation using male rat and hamster liver derived S-9 mix at dose levels of 0.0, 1.0, 3.3, 10.0, 33.0, 100.0, 333.0, 1000.0, 3333.0 µg/plate. The test was repeated and atleast three plates were used at each dose level. Geranyl acetate failed to induce mutation in S. typhimurium TA 1535, TA 1537, TA 98 and TA 100 in the presence and absence of S9 metabolic activation system and hence is not mutagenic in vitro.

 

In a study for structurally and functionally similar read across chemical, Gene mutation toxicity study was performed by D. Wild, M.-T. King et.al. (Fd Chem. Toxic, 1983) to determine the mutagenic nature of Allyl capronate (123-68-2). Gene mutation toxicity was performed to determine the mutagenic nature of Allyl capronate. The test was performed by plate incorporation method using Salmonella typhimurium strains TA98, TA100, TA1535, TA1537, TA1538 with and without S9 metabolic activation system at 5 doses of the test substance were used upto 3600 µg/plate and the plates were incubated for 48hrs. Allyl capronate did not induce gene mutation in Salmonella typhimurium strains TA98, TA100, TA1535, TA1537, TA1538 in the presence and absence of S9 metabolic activation system and hence is not likely to classify as a gene mutant in vitro.

 

Based on the data available for the target chemical and its read across substance and applying weight of evidence 3,7-dimethylocta-2,6-dien-1-yl propionate (105-90-8) does not exhibit gene mutation in vitro. Hence the test chemical is not likely to classify as a gene mutant in vitro.
