Repeated dose toxicity: Oral

Based on the prediction done using the OECD QSAR toolbox version 3.3 with log kow as the primary descriptor and considering the five closest read across substances, repeated dose toxicity was predicted for Pigment Red 81. The study assumed the use of male and female F344 rats in a study for 104 weeks. Since no significant treatment related effects were observed at the mentioned dose level, hence the No Observed Adverse Effect Level (NOAEL) for Pigment Red 81 is predicted to be 370.608337402 mg/Kg bw/day.

Based on this value it can be concluded that the substance is considered to not toxic as per the criteria mentioned in CLP regulation.

Repeated dose toxicity: Inhalation

The test substance Xanthylium, 9-[2-(ethoxycarbonyl)phenyl]-3,6-bis(ethylamino)-2,7-dimethyl-, molybdate tungstate phosphate has very low vapor pressure (4.19E-011 Pa). Also the normal conditions of use of this substance will not result in aerosols, particles or droplets of an inhalable size, so exposure to humans via the inhalatory route will be unlikely to occur and therefore repeated dose toxicity via the inhalation route was considered for waiver.

Repeated dose toxicity: Dermal

The acute dermal toxicity value for Xanthylium, 9-[2-(ethoxycarbonyl)phenyl]-3,6-bis(ethylamino)-2,7-dimethyl-, molybdatetungstatephosphate

(as provided in section 7.2.3) is >2000 mg/kg body weight. The substance was also found to be not irritating and not sensitizing to the skin. Based on these considerations, the end point for repeated dermal toxicity is considered as waiver.

Prediction model based estimation and data from read across chemicals have been reviewed to determine the toxic nature of Pigment Red 81 upon repeated exposure by oral route. The studies are as mentioned below:

The study assumed the use of male and female F344 rats in a study for 104 weeks. Since no significant treatment related effects were observed at the mentioned dose level, hence the No Observed Adverse Effect Level (NOAEL) for Pigment Red 81 is predicted to be 370.608337402 mg/Kg bw/day.

The above mentioned predicted data for the target chemical is futher supported by the data from read across chemicals as mentioned below:

Combined repeated dose repro-devp. Screen was performed by Burnett and Goldenthal (Food and chemical toxicology, 1986) to determine the toxic nature of functionally similar read across chemical D & C Yellow No. 8, sodium fluorescein (RA CAS no 518 -47 -8; IUPAC name: disodium 2-(3-oxo-6-oxidoxanthen-9-yl)benzoate) upon repeated exposure by oral route.An aqueous solution of the dye was administered by garage to groups of 25 Charles River Sprague-Dawley rats at doses of 100, 500 and 1500 mg/kg on days 6- 19 of gestation. A control group received water on a comparable basis. Individual doses were determined on the basis of body weights recorded on days 6, 9, 12 and 16 of gestation.Survival was 100% in the controls and the groups receiving 100 and 500 mg/kg of dye. Six rats in the high-dose group (1500 mg/kg) died during the dosing period. There were slight reductions in body-weight gains in 1500 mg/Kg bw group, compared with controls, throughout the dosing period.Orange discoloration of the urine was noted in all treated rats during the treatment period.At autopsy, green discoloration of the amniotic fluid was noted in 1, 10 and 16 rats in the 100, 500 and 1500 mg/kg/day groups, respectively, and the small intestines were green in colour in many rats in the 500 mg/Kg group. Based on the above observations made, the No Observed adverse effect level (NOAEL) forD & C Yellow No. 8, sodium fluorescein is considered to be 1500 mg/kg bw

14 days repeated dose toxicity study was performed to determine the toxic nature of structurally and functionally similar read across chemical Rhodamine 6G (RA CAS no 989 -38 -8; IUPAC name: 9-[2-(ethoxycarbonyl)phenyl]-3,6-bis(ethylamino)-2,7-dimethylxanthenium chloride). Groups of five males and five females mice were fed diets containing 0, 31, 62, 125, 250, or 500 mg/kgrhodamine 6G for 14 consecutive days. Animals were housed five per cage. Feed and water were available ad libitum. All mice lived to the end of the studies. Mice that received 500 mg/Kg gained little or no weight. The final mean body weights of mice that received 250 or 500 mg/Kg were 8% or 18% lower than that of controls for males and 2% or 8% lower for females. Feed consumption by dosed mice was similar to that by controls. No compound-related clinical signs were observed. Based on these considerations, the No Observed Adverse Effect Level (NOAEL) for Rhodamine 6G is considered to be 500 mg/Kg.
