Details on sampling:
Test concentrations:
In a pretest the LC50 (96 h) was determinated to be > 300 mg/l. Based on the pretest, the test concentrations were chosen as 32/56/100/178 and 316 mg/l (nominal). The pretest was not conducted under GLP conditions, but the results are archivated in the file of the main test. A control was carried out with dechlorinated tap water.

Mode of preparation:
The test substance was by ultrasonic treatment dissolved in 200 ml water, which was slowly added to the fish bath.

Analysis of the test substance:
Water samples were taken from the fish baths at the beginning, after 48 hours and at the end of the experiment. All test medium samples for the analytical determinations were taken after mixing of the test solutions and kept at -18°C to -25°C until analysis. The concentrations of the test substance were analysed by the Central Analytical Dept. The pretest was not conducted under GLP conditions, but the results are archivated in the file of the main test.
Vehicle:
yes
Test organisms (species):
Danio rerio (previous name: Brachydanio rerio)
Details on test organisms:
The test organisms were fish of the species Brachydanio rerio (Zebra fish) supplied by West Aquarium, Bad Lauterberg, Germany.
Prior to the test the fish were held in the laboratories of the Product Ecology TF for 60 days in dechlorinated tap water under similar conditions as in the test. During this time they were regularly fed with Tetramin fish food.
For each test concentration, 10 fish were used.
Fish weight: 2.4 g (mean of all test organisms)
Fish length: 29.3 mm (mean of all test organisms)
Fish age: 125 days
Treatment: none
Test type:
static
Water media type:

Limit test:
no
Total exposure duration:
96 h
Hardness:
179 mg/l CaCO3
Test temperature:
20.2 - 21.7 °C
pH:
7.9 - 8.5
Dissolved oxygen:
86 - 97 %
Nominal and measured concentrations:
Nominal concentration: 32, 56, 100, 178 and 316 mg test substance/I.
Measured concentration: 15, 32.4, 85.1, 125, 195 mg test substance/I.
Details on test conditions:
TEST SYSTEM
Static system, 96 hours at nominal 21 - 25°C (± 1°C during the test), yellow light for 12 hours daily.
The fish were exposed in dechlorinated tap water (carbon filter) in a 5 litre glass aquaria with a volume of 2.96 - 2.92 litre (3,0 litre minus 4x 0,02 Litre for analytical control). During exposure, the animals were kept at 20.2 - 21.7°C, slowly aerated, non feeding.

Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Duration:
96 h
Dose descriptor:
NOEC
Effect conc.:
ca. 195 mg/L
Nominal / measured:
meas. (arithm. mean)
Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
The analytically determined test concentrations of the test substance in the fish baths at the beginning of the test varied between 85.6% and 108.9% of the nominal values. At the end of the test concentrations between 26.6% and 86.0% of the nominal values were found.
Because of this decrease of the test substance during the test the NOEC- and the LC-values are related to the mean values of the measured concentrations at the beginning, after 48 hours and at the end of the test.
During the test no mortality occured in the control and in the test vessels up to the highest concentration. Therefore, the NOEC and LCQ was 195 mg/l. The LC50-values during the test and the end could not be calculated, but were clearly higher than 195 mg/l.

Control:
Mortalities in blank : 0%

Calculation of LC50 - Values:
The LC50-values and the 95% confidence limits could not be calculated.

Responses of the test fish:
No abnormal behaviour of the test fish was observed.
Validity criteria fulfilled:
not specified
Conclusions:
NOEC after 96 h = 195 mg/l;
LC0 after 96 h = 195 mg/l;
LC50 after 96 h > 195 mg/l;
LC100 after 9 6 h > 195 mg/l
Executive summary:

Method:

The short-term toxicity to fish of the test substance was determined according to the OECD Guideline 203 (Fish, Acute Toxicity Test).

Observations:

Due to the results of a pretest the nominal concentrations for the main test were selected as : 32/56/100/178 and 316 mg test substance/l. The test the concentrations in the analysed test media at the end of the test were decreased to 26.6% - 86.0% of the nominal values. Therefore, the reported results are related to the mean analyzed concentrations of the test substance.

Conclusion:

NOEC after 96 h = 195 mg/l

LC0 after 96 h = 195 mg/l

LC50 after 96 h > 195 mg/l

LC100 after 9 6 h > 195 mg/l

Description of key information

NOEC after 96 h = 195 mg/l

LC0 after 96 h = 195 mg/l

LC50 after 96 h > 195 mg/l

Additional information

The short-term toxicity to fish of the test substance was determined according to the OECD Guideline 203 (Fish, Acute Toxicity Test).

Due to the results of a pretest the nominal concentrations for the main test were selected as : 32/56/100/178 and 316 mg test substance/l. The concentrations in the analysed test media at the end of the test were decreased to 26.6% - 86.0% of the nominal values. Therefore, the reported results are related to the mean analyzed concentrations of the test substance.

The LC50 calculated for the test substance in this test was > 195 mg/l.

This result is in line with the one obtained in other tests listed in the table attached below.
