Half life of hydrolysis of test chemical Bis(4-hydroxy-N-methylanilinium) sulphate (CAS no. 55-55-0) is predicted using OECD QSAR tool box v3.3 using log Kow as primary descriptor. The half life of hydrolysis of test chemical Bis(4-hydroxy-N-methylanilinium) sulphate was estimated to be 153.80 days. Since half hife of hydrolysis for this chemical is more than 10 days this chemical is considered to be non hydrolysable.

Biodegradation in water:

Biodegradation experiment was performed in batch system using adapted activated sludge as inoculums. Activated sludge was obtained from sewage plant then it is adapted for 20 days with the test chemical Bis(4-hydroxy-N-methylanilinium) sulphate (CAS no. 55-55-0) in biological medium containing Glucose peptone etc. After activated sludge get adapted concentration of test chemical was increased upto 200 mg/L and then biodegrdation was observed for test chemical. Test chemical was the only source of organic carbon. All adaption process and degradation process was taken place in dark room at 20 oC. Experiment was carried out till there is no decrease of COD. Inoculum blank flask was also set up simultaneously. Sampling was done at definite time interval 50-80 mL sample was taken for analyzing COD. The experiment is carried out till there is no decrease of COD. After that time the total percentage of COD removed (% degradation) and the rate of degradation are evaluated. Percent biodegradability of test chemical Bis(4-hydroxy-N-methylanilinium) sulphate observed to be 59.4 % by COD (Organic Carbon ) and rate of degradation was 0.8 mg COD /g/L per hour. If the degradation rate being over 15 mg of COD g /L/h), the substance tested can be considered to be biologically readily decomposable but for test chemical it was only 0.8mg COD g/L/h. Thus it is concluded that test chemical Bis(4-hydroxy-N-methylanilinium) sulphate is inherently biodegradable.

Using BCFBAF Program (v3.00) model of EPI suite 2017 the estimated bio concentration factor (BCF) for Bis(4-hydroxy-N-methylanilinium) sulphate (CAS No. 55-55-0) is 3.162 L/kg wet-wt at 25 deg. c which does not exceed the bioconcentration threshold of 2000. Therefore it is concluded that test chemical Bis(4-hydroxy-N-methylanilinium) sulphate is non bioaccumulative in food chain.

The Soil Adsorption Coefficient i.e. Koc value of test chemical Bis(4-hydroxy-N-methylanilinium) sulphate (CAS No. 55-55-0) was estimated using EPI suite KOCWIN Program (v2.00) as 830 L/kg (log Koc= 2.9194) by means of MCI method at 25 deg. C. This log Koc value indicates that test chemical Bis(4-hydroxy-N-methylanilinium) sulphate has moderate sorption to soil and therefore have slow migration potential to ground water.

Predicted data for target compound Bis(4-hydroxy-N-methylanilinium) sulphate (CAS no. 55-55-0) and supporting weight of evidence study for its closest read across chemical with log Kow as primary descriptor were reviewed for the Hydrolysis endpoint which is summarized below.

Similarly, in supporting weight of evidence study done from Hazardous Substance Data Bank,( HSDB, 2017) the half life of hydrolysis of read across chemical (2-hydroxy-4-(octyloxy)phenyl)(phenyl)methanone (CAS no. 1843-05-6) was determined by using OECD guideline 111. The half life of hydrolysis of read across chemical (2-hydroxy-4-(octyloxy)phenyl)(phenyl)methanone at pH 4, 7 and 9 determined was greater than 1 year and at 50 oC temperature . Since half life of hydrolysis at pH 4, 7 and 9 is greater than 10 days so it is concluded that read across chemical (2-hydroxy-4-(octyloxy)phenyl)(phenyl)methanone is not expected to undergo hydrolysis.

On the basis of results for target chemical Bis(4-hydroxy-N-methylanilinium) sulphate (CAS no. 55-55-0) (From OECD QSAR tool box v3.3) and for its read across chemicals (from HSDB, 2017) it is concluded that target chemical Bis(4-hydroxy-N-methylanilinium) sulphate is not expected to undergo hydrolysis.

Next supporting weight of evidence study done from Hazardous Substance Data Bank (HSDB, 2017) of read across chemical 4-(methylamino)phenol (CAS no. 150-75-4) and BCF value determined was 3.2 dimensionless by using log Kow of 0.79 and regression derived equation.

Another supporting weight of evidence study done from authoritative database (J check, 2017) in this study Bioaccumulation test was conducted for 6 weeks for determination the bioconcentration factor (BCF) of read across chemical 4-(anilino)phenol (CAS no 122-37-2) on test organism Cyprinus carpio. Nominal concentrations used were 1st Concentration area: 0.1 mg/L (w/v), 2nd Concentration area: 0.01 mg/L (w/v) and Range finding study was carried out on Rice fish (Oryzias latipes) LC50 (48h) 0.714 mg/L. The vehical used in this study was HCO-40, olive oil. The lipid content of fish at start of exposure was 4.2%. Thus according to flow through Fish Test the BCF value for read across chemical 4-(anilino)phenol was observed to be to be in range 3.3-9.2 dimensionless at dose concentration 0.1 mg/L and 14-49 dimensionless at dose concentration 0.01 mg/L on test organism Cyprinus carpio during 6 weeks period.

On the basis of above results for target chemical Bis(4-hydroxy-N-methylanilinium) sulphate (CAS No. 55-55-0) (from EPI suite 2017) and for its read across chemicals (From HSDB and J check) it can be concluded that the BCF value of test substance Bis(4-hydroxy-N-methylanilinium) sulphate is 16.26 L/Kg which does not exceed the bioconcentration threshold of 2000, indicating that the chemical Bis(4-hydroxy-N-methylanilinium)sulphate is expected to be non bioaccumulative in the food chain.
