Description of key information

The NOEC based on mortality was the 25% saturated solution (0.033 mg dehydroabietyl alcohol/L, converted to 0.16 mg hydrogenated rosin alcohols/L based on a dehydroabietyl alcohol content of 20.889%). For reproduction (number of offspring per surviving Daphnia) and length of parent Daphnia, the NOEC was the 50% saturated solution.

A Daphnia reproduction study was conducted according to OECD 211, with Daphnia magna (Scymaris 2021). Hydrogenated rosin alcohols is a UVCB with four main constituent groups and as the substance is difficult to test, method development work was carried out prior to the study in order to determine the most appropriate way of preparing test solutions. The use of saturator columns was found to offer the most consistent results and was therefore used in the definitive study. A semi-static test was conducted, with test solutions prepared prior to each renewal using the saturator columns (<24 hour collection periods) and renewal of test solutions three times a week. Test concentrations of 100, 50, 25, 12.5 and 6.25% of the saturated solution were included in the study, alongside a control (dilution water) and saturator column control (column dosed with acetone but no test item and otherwise prepared in the same way as the test solutions).

Test item concentrations were monitored using GC-MS. The analytical results, based on the dehydroabietyl alcohol constituent, showed that results for the freshly prepared β€˜on’ solutions were close to the expected solubility for this constituent, with some reduction over the renewal periods. Results are based on time weighted average concentrations. 

Significant differences were observed for mortality in the 100 and 50% saturated solutions. The NOEC based on mortality was therefore the 25% saturated solution (0.033 mg dehydroabietyl alcohol/L, converted to 0.16 mg hydrogenated rosin alcohols/L based on a dehydroabietyl alcohol content of 20.889%). For reproduction (number of offspring per surviving Daphnia) and length of parent Daphnia, the NOEC was the 50% saturated solution.
