Description of key information

Carcinogenicity of 2-Benzyl-4-(dimethylamino)butanoic acid was estimated by using the following predictors: VEGA (three models), Derek Nexus and Toxtree.

The predictors were employed in order to apply a weight of evidence (WoE) approach to enhance the reliability of the prediction. In the weight of evidence assessment only reliable predictions are to be taken into account. Vega predicted the target compound as: NON-Carcinogen (Low reliable - CAESAR model), Carcinogen (Moderate reliable - ISS model), Possible NON-Carcinogen (Moderate reliable - IRFMN/Antares model) and Possible NON-Carcinogen (Moderate reliable - IRFMN/ISSCAN-CGX model). Derek Nexus identified no alerts for the target compound.

Toxtree predicted the target compound as negative for genotoxic carcinogenicity but found a structural alert for nongenotoxic carcinogenicity.

Thus, it was concluded that 2-Benzyl-4-(dimethylamino)butanoic acid was predicted as positive for carcinogenicity but theprediction was assessed as low reliable.

Additional information

The prediction was provided on the substance 2-Benzyl-4-(dimethylamino)butanoic acid (CAS 1613 -23 -6), the acid form of the substance in the object of the registration.
